Objectives: The differentially expressed proteins were identified from the earlier stage of the research group in the process of gastric carcinogenesis.The clinical significance of expression of Annexin A1 and SOD3 proteins were analyzed in gastric carcinogenesis.The results will provide a theoretical basis for identifying the proteins used in early diagnosis of gastric cancer.Methods: 1.The expression of Annexin A1 and SOD3 proteins was observed by immunohistochemical staining combined with tissue microarray including 234 cases of gastric adenocarcinoma,86 cases of metastatic lymph node carcinoma,142 cases of normal gastric mucosa and 217 cases of adjacent tissues in different stages of gastric carcinogenesis.Combined with clinical data,the relationship and significance between the two proteins expression were analyzed in gastric mucosal carcinogenesis and related clinical data.ROC curve and immunohistochemical staining were used to analyze the value of Annexin A1 and SOD3 protein expression in the diagnosis of gastric mucosal carcinogenesis.Results:1.The result of immunohistochemical staining showed that Annexin A1 protein was mainly expressed in cytoplasm,and the positive rates of expression were respectively 0.70%(1/142),1.84%(4/217),14.10%(33/234)and 33.72%(29/86)% in normal gastric mucosa,paracancerous tissues,gastric cancer tissues and lymph node metastatic cancer tissues.Annexin A1 protein was highly expressed in gastric adenocarcinoma and lymph node metastasis,but was lower in normal gastric mucosa and paracancerous tissues.The expression of Annexin A1 protein was higher in paracancerous tissues than in normal gastric mucosa tissues,and gradually increased in various stages of gastric mucosal carcinogenesis(P<0.05).In addition,the positive rates of Annexin A1 protein expression were respectively 9.64%(8/83),13.4%(15/112)and 25.64%(10/39)in well-differentiated adenocarcinoma,moderately differentiated adenocarcinoma and poorly differentiated adenocarcinoma.The expression of Annexin A1 was negatively correlated with the differentiation of gastric adenocarcinoma,but positively correlated with TNM stage and lymph node metastasis of gastric carcinoma.The lower the differentiation of gastric adenocarcinoma was the higher the positive rate of Annexin A1 protein expression,and the positive rate of Annexin A1 protein expression was high in gastric adenocarcinoma after lymph node metastasis(P<0.05).2.The result of immunohistochemical staining showed that SOD3 protein was mainly expressed in cytoplasm,and the positive rates of expression were respectively 91.55%(130/142),71.43%(155/217),29.49%(69/234)and 18.60%(16/86)% in normal gastric mucosa,paracancerous tissues,gastric cancer tissues and lymph node metastatic cancer tissues.SOD3 protein was lowerly expressed in gastric adenocarcinoma and lymph node metastasis,but was higher in normal gastric mucosa and paracancerous tissues.The expression of SOD3 protein was lower in paracancerous tissues than in normal gastric mucosa tissues,and gradually decreased in various stages of gastric mucosal carcinogenesis(P<0.05).In addition,the positive rates of SOD3 protein expression were respectively 38.55%(32/83),26.79%(30/112)and 17.94%(7/39)in well-differentiated adenocarcinoma,moderately differentiated adenocarcinoma and poorly differentiated adenocarcinoma.The expression of SOD3 was positively correlated with the differentiation of gastric adenocarcinoma,but negatively correlated with TNM stage and lymph node metastasis of gastric carcinoma.The lower the differentiation of gastric adenocarcinoma was the lower the positive rate of SOD3 protein expression,and the positive rate of SOD3 protein expression was low in gastric adenocarcinoma after lymph node metastasis(P<0.05).3.The sensitivity of Annexin A1 and SOD3 protein expression was respectively 28% and 54% to distinguish normal gastric mucosa from gastric adenocarcinoma.The specificity for normal gastric mucosa and gastric adenocarcinoma was respectively 32% and 56%.The sensitivity and specificity of the combined detection of Annexin A1 and SOD3 were respectively 65% and 72% in the differential diagnosis of normal gastric mucosa and gastric adenocarcinoma.The sensitivity and specificity were both increased to combined detection of Annexin A1 and SOD3 proteins expression,which was more significant than that of single identification of normal gastric mucosa and gastric adenocarcinoma.Conclusions: Annexin A1 protein is highly expressed in gastric cancer tissues and SOD3 protein is low expressed in gastric cancer tissues.Two expression is related to the hypholog types,TNM staging and lymph node metastasis of gastric cancer.The combined detection of Annexin A1 and SOD3 proteins is of more clinical significance in the diagnosis of gastric cancer.
