| Objective To observe the effects of herbal-cake-separated-moxibustion on the expression of Ghrelin and VIP in functional gastrointestinal disorders" modal rats with syndrome of liver stagnation and spleen deficiency, and to explore the possible mechanisms of herbal-cake-separated-moxibustion in treating functional gastrointestinal disorders.Methods60healthy SD rats were randomly divided into5groups after three-day’s keeping in different cages according to sexes. They were blank group (Group A), model group (Group B), herbal-cake-separated-moxibustion group (Group C), domperidone group (Group D) and Xiaoyaosan group (Group E),12rats per group. Except for the blank group, the other4groups underwent modelling by chronic constraint, irregular diet, excessive fatigue, clamping tails and shaking lasting for three weeks. Then treated those model rats with different therapies according to the group it belongs to for two weeks. Namely, the blank group were felt normally; the model group underwent bind for30minutes then gave normal saline; the herbal-cake-separated-moxibustion group underwent herbal cake separated moxibustion for30minutes then gave normal saline; the domperidone group and Xiaoyaosan group underwent bind for30minutes then gave domperidone and Xiaoyaosan respectively with the same dosage as group B. After that, all the rats were given nothing for24hours, killed them by anesthesia, and get their blood and other organs, including gastric antrum and cord. Then measured the levels of serum VIP and Ghreelin. Get the data and analysed them.Results:1, General situation:Before modeling, all of the rats were active and in a good mental state. Their fur were glossy and their excrement were spherical, neither dry nor wet. After modeling, the model rats became tired, fidgety and in a bad mental state. Their fur get messy and yellow, and their excrement get watery. After treatment, the situation of the rats in the herbal-cake-separated-moxibustion group, domperidone group and Xiaoyaosan group get better.2, Body weight and food intake: After modeling, the rats’ appetite decreased and the weight growth get slows, the difference was statistically significant (P<0.05,P<0.01).3, Mine field experiment:After modeling, the rats’ activities decreased (P<0.05,P<0.01).4, The rate of serum VIP:Compared with the blank group, the rate of serum VIP in the model group was higher (P<0.05) Compared with the model group, the rates in the herbal-cake-separated-moxibustion group, domperidone group and Xiaoyaosan group decreased (P<0.05,P<0.01).5, The expression of Ghrelin in the tissue of gastric antrum and cord:Compared with the blank group, the expression of Ghrelin in the tissue of gastric antrum and cord in the model group decreased (P<0.05,P<0.01); while, the expression in the herbal-cake-separated-moxibustion group, domperidone group and Xiaoyaosan group were lower than that of in the model group (P<0.01)Conclusion:Herbal-cake-separated-moxibustion can reduce the rate of SD rat’s serum VIP and increase the expression of Ghrelin in the tissue of gastric antrum and cord. It might be one of the possible mechanisms of herbal-cake-separated-moxibustion in treating functional gastrointestinal disorders. |
