| Object: To observe the influence of cake-separated moxibustion for the absorption and paresthesia in rats with FGIDs (liver depression and spleen deficiency syndrome), the influence of5-HT, DA neurotransmitters and the expression and activation of AMPA receptor subunits in the cell of the hippocampus and amygdala, and to explore the possible mechanism of cake-separated moxibustion for treating FGIDs (liver depression and spleen deficiency syndrome) rats, so it can provide some experimental basis for the clinical application of cake-separated moxibustion with FGIDs.Methods:1. Rats with liver depression and spleen deficiency syndrome as the research object.48SD rats were randomly divided into4groups according to the table of random number method:Blank group, Model group, cake-separated moxibustion group, Xiao-yao-san group. Observation the rat behavior, the signs and symptoms, detection the rate of urinary D-fructose excretion and intestinal sensitivity test. Using high efficiency liquid, Real time-PCR and other technical means to detected the expression of colon5-HT, DA and hippocampus, amygdala c-fos.2.72SD rats were randomly divided into6groups:blank group, model group, Cake-separated moxibustion group, Cake-separated moxibustion+CNQX group, CNQX group, sham group. Using Western blot, Real time-PCR and other technical means, a cake-separated moxibustion for intervention, in the amygdala trace injected AMPA receptors antagonist CNQX for comparison, to observe the expression changes of central hippocampal CA1, CA3and DG and amygdala BLA GluR1and GluR2.Result:1. The model rats for the first time against disturbing, gradually show the cluster or lack of exercise, an air of fatigue, hair brown scattered dull drooping beard, loose stool, etc. he rats weight and appetite slow increased, the cross number, stand number and modified number decreased significantly; the rate of the urinary excretion of D-fructose excretion had continued to reduce, but bowel sensitivity increased. Compared with the model group, the cake-separated moxibustion group and XiaoYaoSan group revealed that urinary D-fructose excretion was increased and the intestinal sensitivity was reduced, the difference was statistically significant (all P<0.01).2. Compared with the blank group, the expression of the rats5-HT and DA in the colon tissue increased in the model group, the difference was statistically significant (P<0.01or P<0.05). Compared with the model group, the expression of the rats5-HT and DA in the colon tissue decreased in the Xiaoyaosan group and Cake-separated moxibustion group, the differences were statistically significant (P<0.05or P<0.01). Compared with the Xiaoyaosan group, the1Cake-separated moxibustion group difference was statistically significant (all P<0.05). In the hippocampus, amygdala, compared with the blank group, the expression of c-fos in the model group were statistically significant (all P<0.05). Compared with the model group, the Cake-separated moxibustion group and the Xiaoyaosan group were statistically significant (all P <0.05). The Cake-separated moxibustion group and the Xiaoyaosan group had statistically significant (P<0.05). 3. In the hippocampal CA1region, CA3region and Amygdala BLA region, compared with the normal group, the expression of the GluRl and GluR2of the model group and the sham operation group were statistically significant (all P <0.05). Compared with the model group, the Cake-separated moxibustion group, the Cake-separated moxibustion+CNQX group and CNQX group differences were statistically significant (all P<0.05). Compared with the CNQX group, the expression of the GluR2of the Cake-separated moxibustion+CNQX group were statistically significant (P<0.05).In the DG region of the hippocampus, compared with the blank group, the expression of the GluR2of the model group and the sham operation group were statistically significant (all P<0.05).4. Groups of rats with gastric mucosa and knot bowel wall epithelium structure was integrity, the cells arranged orderly, submucosa and muscular was clear, part of the mucous membrane of model group and sham-operation group were occasional vasodilation, occasional see few inflammatory cells.Conclusion:1. Using the composite etiology modeling method successfully to cause the FGIDs rats (liver stagnation and spleen deficiency syndrome model), Cake-separated moxibustion can promote the gastrointestinal absorption function obviously, improve the visceral sensitivity, which suggest that improving the gastrointestinal absorption and visceral sensitivity maybe is one of the effectively parts about Cake-separated moxibustion treatment the FGIDs.2. Cake-separated moxibustion can decrease the expression of5-HT and DA in the colon, c-fos in central hippocampus and amygdale, which suggest that the brain gut axis-central avenue maybe the key parts about Cake-separated moxibustion on the FGIDs. 3. The "excited-inhibition" imbalance of the hippocampus and amygdala of the rats with FGIDs (the Liver depression and spleen deficiency syndrome), antagonistic amygdala area AMPA receptors, inhibited amygdala excited, this may be an important way for the cake-separated moxibustion regulating the "excited-inhibition " imbalance of the amygdala and hippocampus. |
