| AIM:To observe the effect of cake-separated-moxibustion on the expression of mineralocorticoid receptor(MR) and glucocorticoid receptor(GR) in hippocampal of liver stagnation and spleen deficency model rats with functional dyspepsia(FD).To explore the possible mechanism of cake- separated- moxibustion with FD throuth the Limbic system-Hypothalamic-Pituitary-Adrenal axis.METHODS: Fifty SD rats were divided into five groups according to the random number table: blank group(A), model group(B), cake-separatedmoxibustion group(C), moxibustion group(D) and Xiaoyaosan group(E), 10 rats in each group. In addition to group A, the other four groups were made with chronic restraint stress + excessive fatigue + irregular food for 21 days. After the molding,group B was bounded for 30 minutes and given 0.9%NaCl by gavage;group C was bounded for 30 minutes、underwent herbal cake-separated- moxibustion and given0.9%NaCl by gavage;group Dwas bounded for 30 minutes 、underwent moxibustion and given 0.9%NaCl by gavage;while group E was bounded for 30 minutes and given Xiaoyaosan by gavage for the same duration, respectively. The intervened treatment last for 14 days. The general condition and behavior of all rats were observed during the experiement.At the end of the experiment, the gastric residual was observed and the expression of MR and GR in hippocampal were measured by Reverse Transcription-Polymerase Chain Reaction(RT-PCT).RESULTS:1. After modeling,the rats showed tired, fretful, fur dim and withered,their bowels were either drier or more watery; the number of grid crossing、standing up and modifing were significantly reduced than before(P < 0.01,or P < 0.05).2. After the treatment, the rats’ general conditions improved: their air was much soffer and lustered,their appetite increased 、stool gradually formed and the number of climing and modifying were increased gradurally.3.After the treatment, The gastric residual rate in group B was obviously lower than that of group A(P <0.01), while the gastric residual rate in three treatment groups were higher than that of group B(all P<0.01), but there were no significant differences among three treatment groups(P > 0.05).4.After the treatment, the expression of MR in hippocampal in group B was statistically alike to group A(P > 0.05), while the expression of GR was significantly lower than that of group A(all P < 0.05). Compared to group B, the expression of MR in hippocampal in Group C, D, and E were statistically the same(P>0.05), but the expression of GR were significantly higher(all P < 0.05). There were no significant differences in the three treatment groups,although the expression of GR in group C was much higher(P > 0.05).CONCLUSION: Cake-separated-moxibusiton can increase the gastric residual rate to improve the motivation of gastrointesine of FD rats.It also can reaise the expression of GR in hippocampas to accommodate the function of limbic system hypothalamic pituitary adrenal axis(LHPA/HPA axis). It might be one of the important mechanisms of FD treated by cake-separated-moxibusiton. |
