Semen holarrhenaeb, dry seeds of Holarrhena antidysenterica Wall.ex A.DC, is thecommon drug of Mongolian medicine, which has high value and special effects on flu,hepatitis and enteritis. In recent years the research on Mongolian medicine has been donewidely, but there is none of reports of research on Semen holarrhenaeb. In this text, the initialresearch was conducted on a kind of alkaloid-one of active products of Semen holarrhenaeb,and made a foundation to further research to active products of Semen holarrhenaeb.Ethanol extraction was used to extract active products of Semen holarrhenaeb, and theanalysis was made to ethanol concentration, extracting temperature, and extracting time. Anorthogonal test design was applied to confirm the optimum condition of extracting the totalalkaloids. Organic solvent extraction and silic gel chromatography was used to purify activeproducts, and monomeric alkaloids could be gained. Finally studies were done onantibacterial effects of crude extract and total alkaloids.The results showed that the best technology condition was as follows: extractingtemperature for60℃, extracting time for6h, ethanol concentration for70%. The extractionyield of Crude extracts can be up to5.36%by the process.monomeric alkaloids was gainedafter organic solvent extraction silic gel chromatography. Its crystal purity was94.89%determined in high efficiency liquid chromatography and monomeric alkaloids wasdetermined as conessine by mass spectrum and nuclear magnetic resonance. The antibacterialeffects of crude extract and total alkaloids were studied with six kinds of bacterium whichwere Escherichia coli ATCC25922, Sarcina lutea AS1.880, Corynebacterium pekinenseAS1.29, Escherichia coli AS1.543, Staphylococcus aureus AS1.72, and Listeriamonocytogenes CMCC34005. The ethanol extracts of Semen holarrhenae had evidentlyantibacterial effect to common bacterium, so it can be deduced that70%ethanol extractscontain active component to antibacterial. The minimum inhibitory concentration of crudeextract from Semen holarrhenae to Escherichia coli AS1.543, Escherichia coli ATCC2592,Staphylococcus aureus AS1.72and Sarcina lutea AS1.880are100mg/mL. The minimum inhibitory concentration of total alkaloids to Staphylococcus aureus AS1.72, Escherichia coliAS1.543, Listeria monocytogenes CMCC34005is50,75,25mg/mL. The total alkaloids havestronger antibacterial effects than crude extract. It can be deduced that total alkaloids fromSemen holarrhenae have strong antibacterial effects on bacterium.
