The chemical composition of traditional Chinese medicine is a very complex material system,especially alkaloid compounds.The complexity and diversity of alkaloids make the separation and purification extremely difficult,which has always been a major problem in the basic research of traditional Chinese medicine.Aiming at the difficulties in the separation and purification of alkaloid compounds,a high performance liquid chromatography(HPLC)method was used to separate and characterize alkaloids from Chelidonium majus L.in this thesis.Aiming at the difficulties in separation of alkaloid compounds,a two-dimensional offline reversed-phase chromatographic method was constructed,and the developed two-dimensional separation system was applied to the separation and analysis of alkaloid compounds in Chelidonium majus L..Large-scale separation of alkaloid compounds in Chelidonium majus L.was achieved through optimization of experimental conditions and linear amplification.Then,mass spectrometry and nuclear magnetic characterization were used to identify the structure of alkaloids separated from Chelidonium majus L..11 alkaloid compounds were separated from the total alkaloids of Chelidonium majus L.,and(S)-N-Methylstylopine and Noroxyhydrastinine were the first separation from Chelidonium majus L..The experiment shows that the two-dimensional offline reversed-phase chromatographic separation system has realized the efficient separation of alkaloid compounds in Chelidonium majus L..Aiming at the problems of long time and heavy workload of traditional compound identification methods such as nuclear magnetic characterization,a method was developed for the rapid and systematic identification of Chelidonium majus L.alkaloids by HPLC-Q-TOF/MS.By analyzing the mass spectrometry data of different types of alkaloids,the mass spectrometry of Chelidonium majus L.alkaloids was summarized by combined with relevant literature,the alkaloid compounds in Chelidonium majus L.were systematically identified and verified with a reference substance.21 alkaloid compounds were identified from the total alkaloid extracts of Chelidonium majus L.,(S)-N-methylstylopine was first reported in Chelidonium majus L.and dihydrocoptisine and norchelidonine were identified for the first time in Chelidonium majus L.This method was fast,accurate and efficient,and has important significance for the preliminary qualitative research of the components in the complex system of natural products and the separation and purification of alkaloids.This thesis establishes a method for the separation of alkaloids by two-dimensional offline reversed-phase liquid chromatography,which realizes the efficient separation of alkaloids in Chelidonium majus L.and identification of alkaloid compounds by liquid chromatography-mass spectrometry(LC-MS).It has certain promotion effect on understanding the quality foundation of Chinese medicine and improving the separation and purification level of Chinese medicine.
