Common and frequently encountered in otorhino laryngology, Pharyngitis is inflammation inpharyngeal mucosa and lymphoid tissue. The study confirmed that YinhuaQingyan tablet has a good effect on Pharyngitis. It is a kind of prescription used in clinical and mainly cure acute or chronic Pharyngitis. Clinical practices for many years have shown this prescription has a quick and acurate effect. It can evidently improve the symptom, shorten the treatment cycle and have no apparent toxic and side-effect. So, it is popular with the patients. This study based on the clinical practice and applied modern preparation technology concerns three aspects:preparation, quality standard and pharmacodynamics. The prescription can be made of quick effect tablets, which will be convinient for patients and improve the compliance.By using water extraction the prescription has high viscosity and is not easy to dry, and it is made under reduced pressure and wet granulation. To ensure the efficacy of case with excipients as little as possible, the experiments on its amount were investigated. Starch, dextrin, lactose is a diluents prescription in the accessorces. The test found that under the condition of the total volume of the same case, the best ratio of diluent starch, dextrin, lactose, were examined to shorten disintegration time. By orthogonal experiments, the amount of disintegrants, lubricants, wetting agents were investigated. The optimal prescription was at the temperature of60℃, relative density being (1.2to1.3) prescription medicine extract added to the diluent of extract quality twice, including starch, dextrin, lactose mass ratio of3:1:1. The amount of6%of the total weight of particles in the whole. Sodium carboxymethyl starch was selected as a disintegrating agent. The amount of the total weight of particles in the whole is1%in choice of magnesium stearate as a lubricant and50%ethanol as a wetting agent.The optimised prescription is reasonable, stable and reproducible and has a good effect on disintegration.To ensure the quality of preparation, the active ingredients were examined in the prescription by HPLC, that is. HPLC method was established for the determination of chlorogenic acid, galuteolin and forsythin in yinhua qingyan tablets. Venusil MP C18(4.6mm×250mm,5μm) was employed to determine the content of chlorogenic acid, the mobile phrase was Acetonitrile:0.4%phosphoric acid (10:90, v/v), the detected wavelength was327nm and the temperature was25℃. Forsythin in Fructus forsythiae suspensae was seperated by using Acetonitrile:water (24:76, v/v)as mobil phrase with the flow rate being1mL·min-1when the column temperature was30℃at277nm. Galuteolin was detected by Agilent ZORBAX SB-phenyl column (4.6mm×250mm.5μm) with acetonitrile as mobil phrase A and0.5%glacial acetic acid as mobil phrase B. the gradient elution procedure was0min (A:10%. B:90%)â†'15min (A:18%, B:82%)â†'30min (A:18%. B:82%)â†'40min (A:30%, B:70%)â†'45min CA:40%, B:60%) the flow rate was1mL·min-1. the detected wavelength was350nm and the temperature30℃. The good linear relationship for the chlorogenic acid, galuteolin and forsythin was0.3~2.4μg.0.012~0.096μg and0.26~2.08μg respectively, the average recovery was99.96%.100.30%and100.13%. the relative standard deviation was1.43%.1.75%and1.22%(n=6). This method is reproducible, accurate and specific and can be used for the determination of chlorogenic acid, galuteolin and forsythin. It provides a simple and reliable method to control the quality of yinhua qingyan tablets.According to the clinical diagnostic criteria for acute pharyngitis, the animal models of rats with ammonia throat spray were established Experiments in Wistar rats, half male and half female, were randomLy divided into seven groups:control group, model group, positive medicine group, Yinhua Qingyan tablets of high, medium and low dose group, and spontaneous recovery group. After spraying administration4d modeling, the changes of animal blood indicators and pharynx organizatiions were inspected. The experimental results showed that the Yinhua Qingyan tablets could reduce acute pharyngitis symptoms in rats. Routine blood test showed that each index value tended to be normal. The biopsy revealed acute pharyngitis, phayyngeal tissue morphology in rats improved tissue lesions can be corrected. Seen this light the Yihua Qingyan tables are effective in the treatment of acute paryngitis.
