Objective:To investigate the TCM prescription laws of IUD leeding side responsesand utetine VSMC’s apoptosis of model rats concerning control of Gongning Granula.Methods:1. TCM prescription regular research: To collect the literatures duringJanuary2002~December2012that concerning the TCM prescription medication oftreating IUD bleeding side effects, then use statistical methods to investigate the TCMprescription laws of IUD bleeding side responses.2. Animals experiment: To create IUDcontraceptive rat model with non-invasive method and invasive method, then observe theIUD insertion success rate and expulsion rate, as well as the contraceptive prevalence rate.To collect rats’ peritoneal washings and detect the content of tumor necrosis factor-α(TNF-α), then make comprehensive evaluation of the two modeling method;50female ratswere randomly divided into GongNing Granules group,Indomethacin group, noninvasivemodel group, noninvasive sham group and normal control group, then using noninvasivemethod to establish IUD model rats and give drug intervention, detect the p53and c-Mycprotein expression of uterine vascular smooth muscle cells(VSMC)in rats, to clearGongNing Granules on uterine VSMC apoptosis mechanisms of regulation and controlfunction.Results:1. Prescription law research: The total frequency of traditional Chinesemedicine are512times,of which tonify deficiency of medicine accounted for37.69%,occupies the first place; followed by heat-clearing drugs accounted for16.60%; hemostatic medicine accounted for15.82%; blood-activating and stasis-resolvingmedicinal accounted for13.08%and other drugs accounted for16.81%. By the analysis ofstatistics principle that commonly used medicine for prescription for the treatment of IUDbleeding are Astragalus, Leonurus, Angelica, Radix Rehmanniae, Cattail Pollen,Rubia,Rhizoma Cyperi, Cortex Moutan, Notoginseng, Rhizoma Ligustici Wallichii,Sanguisorba.2. Animal experimental studies: the IUD placed success rate andcontraceptive rate of both noninvasive model group rats and laparotomy model group are100%, the loss rate in noninvasive model group is10%which laparotomy model group is20%, by Fisher’s exact probability method comparison, P=1.000, there is no significantdifferences(P>0.05), but abdominal rinses the TNF alpha of noninvasive model group isobviously lower than laparotomy model group, by comparing both the statistical, thedifference has significant meaning(P<0.01). Noninvasive model group rats’ uterus VSMCof p53and c-Myc protein are higher than in normal group, the difference has significantmeaning(P<0.01). GongNing Granules group and Indomethacin model group’s p53andc-Myc protein expression are significantly lower than that of noninvasive model group(P<0.01). Including GongNing Granules group’s p53and c-Myc protein expression aresignificantly lower than the indomethacin group, two groups through statistical comparisonhave significant difference (P <0.05).Conclusion:1. Modern Chinese medicine treatment of IUD bleeding side effectscommonly use tonic medicine, antipyretic, hemostatic and blood circulation drugs.Commonly prescribed medications contain Astragalus, Leonurus, Angelica, RadixRehmanniae, Cattail Pollen, Rubia, Rhizoma Cyperi, Cortex Moutan, Notoginseng,Rhizoma Ligustici Wallichii, Sanguisorba.2. Using noninvasive method established anIUD contraceptive rats model in an IUD off rate, success rate of placement andcontraceptive use by method of equivalent to laparotomy method. And noninvasive methodin the rats model of inflammatory reaction caused by process and method of the harm ofrats was significantly less than laparotomy, which is more accord with clinical ring features,is an ideal method to made an IUD mold.3. The IUD rats models have uterineabnormalities of VSMC apoptosis mechanism tip cell apoptosis may be one of an IUD bleeding adverse events in the pathogenesis. GongNing Granules can inhibit apoptosis ofuterine VSMC cell through the regulation of p53and c-Myc gene that promptingGongNing Granules can reduce apoptosis of VSMC cell uterine to lower vascular damage,which can protect blood vessels of the uterus to treat the IUD bleeding side effects.
