Emilia Sonchifolia (Linn.) DC belonging to Asteraceae is a kind of medicinal and edible plants, which is commonly used in southern China. There are around100species worldwide, mainly distributed in tropical Asia and Africa, minority found in America. In china it mainly grows in Guangxi, Guangdong, Fujian, Guizhou,Yunnan, Jiangxi, etc. They commonly growth in the hillsides, the fields and roadsides. In China, Emilia Sonchifolia as a medicinal and edible mountain vegetables to be known by the people. It tastes mild and coll. Indications:diarrhea, fever, sore throat, mouth ulcers, pnuemonia, mastitis, enteritis, acute tonsillitis, cholecystitis, upper respiratory tract infections in children, bronchopneumonia, etc. In addition, it is the main raw material of the bonus film and xingpiyanger granules.In this paper, the polysaccharide and the alkaloid extraction process of Emilia Sonchifolia (Linn.) DC were studied. The research of Polysaccharide antibacterial activity to measure the minimum inhibitory concentration (MIC) of the polysaccharide. In addition, making the alkaloids segmentation according to the order of pH and Polarity from small to large. And done the antitumor activity test of all alkaloids. There are few studies content areas:1. Study on extraction technology of polysaccharide from Emilia Sonchifolia.In this paper, Emilia Sonchifolia (Linn.) DC was extracted by water. Study on impact of extraction rate by temperature, solid-liquid ratio, extraction number and extraction time, and then the single factor optimized conditions were obtained:the temperature is80℃, the solid-liquid ratio is1:30, extraction number is2times, extraction time is2h. Further, Get the the optimum conditions by orthogonal of4factors and3levels:the temperature is70℃, the solid-liquid ratio is1:30, extraction number is2times, extraction time is2h. After verifing the optimum condition, the extraction rate is3.38%.2. Senkirkine separation.By use of ethanol extraction and acid treatment in order to obtain the total alkaloids, and then extracted it with dichloromethane. Using silica gel column elution, meantime thin layer chromatography(TLC) trace to detection each gradient elution parts. Selecting acetate-methanol(2:1) elution site, through semi-preparative high performance liquid chromatography for further separation. Then using spectral analysis determine the structure of the monomer compound. Finally, determining the substance is senkirkine.3. Study on extraction technology of alkaloid from Emilia Sonchifolia. Using the isolated senkirkine as the reference substance, and then making the standard curve with analytical high performance liquid chromatography. The process of the ethanol extraction Emilia alkaloids is optimized. Study on impact of extraction rate by ethanol concentration, temperature, extraction time and extraction number. Finally, the single factor optimized conditions were obtained: the ethanol concentration is85%, the temperature is85℃, extraction time is3h, extraction number is2times. Further, Get the optimum conditions by orthogonal of4factors and3levels:the ethanol concentration is85%, the temperature is85℃, extraction time is2h, extraction number is3times. After verifing the optimum condition, the extraction rate of alkaloids is0.87mg/g.4. The study of polysaccharide antibacterial activity.According to the optimum condition of the polysaccharide extracted, taking antibacterial activity test of polysaccharides, and measuring the minimum inhibitory concentration (MIC) of the polysaccharide. Conclusions: polysaccharide inhibit bacillus subtilis, and the minimum inhibitory concentration (MIC) is25mg/mL.5. The study of alkaloid antitumor activity.Making the alkaloids segmentation according to the order of pH and Polarity from small to large. Done the antitumor activity test of all alkaloids. Conclusions:when in the high concentrations, all alkaloids inhibit the cell lines of HepG2except the alkaloid of pH8. When in the low concentrations, only the alkaloid of pH7inhibit the cell lines of HepG2. When in the high concentrations, the alkaloids of pH5, pH7and pH9inhibit the cell lines of Hep3B and resistant cell lines of R-HepG2.
