Emilia sonchifolia (L.) DC, an annual herb, known as "Yi Dian Hong" or "Zi Bei Cao" in Chinese, is usually found in south China. In folk medicine, E. sonchifolia has been used for the treatment of inflammation, rheumatism, dysentery, urinary infections, etc.. The herb has been mainly used to treat gynecological inflammation in clinical treatment, and it’s one of raw materials of Huahong capsule, Huahong tablet and so on. However, there were a small number of studies about the chemical consitituents of this plant, and rare methods were established to evaluate its quality at the present time. In order to use and develop this herb further, the chemical constituents and quality control were studied systematically.Our research results were as follows:1. The chemical compositions of the aerial parts of E. sonchifolia were studied. Thirty-eight chemical constituents were isolated and purified from the aerial parts of E. sonchifolia by silica gel, MCI gel, ODS, Sephadex LH-20, and RP-HPLC column chromatographies. And thirty-six constituents were identified on the basis of physicochemical properties and various spectral data. Those compounds were identified as follows:2-{4-hydroxy-7-oxabicyclo [2.2.1] heptanyl}-acetic acid (1)*,2-(1,4-dihydroxy cyclohexanyl)-acetic acid (2), succinic acid (3), fumaric acid (4), p-hydroxybenzoic acid (5),4-hydroxy isophthalic acid (6), caffeic acid (7),3,4-dihydroxycinnamic acid (8), methyl4-hydroxybenzeneacetate (9), chlorogenic acid (10), methyl chlorogenate (11), rhamnetin (12), isorhamnetin (13), quercetin (14), luteolin (15), tricin7-O-β-D-glucopyranoside (16),8-(2"-pyrrolidinone-5"-yl)-quercetin (17), quercetin-7-O-α-L-rhamnopyranoside (18), isorhamnetin-3-O-α-L-rhamnopyranoside (19), afzelin (20),5,2’,6’-trihydroxy-7,8-dimethoxyflavone-2’-O-β-D-glucopyranoside (21), 5,2’,6’-trihydroxy-7-methoxyflavone-2’-O-β-D-glucopyranoside (22), quercetin-3-O-α-L-rhamnopyranoside (23), isorhamnetin-3-O-rutinoside (24), diosmin (25), linarin (26), quercetin-7-O-β-D-glucopyranoside (27), vitexin (28), vicenin-2(29), esculetin (30), isowedelolactone (31), brevifolin (32), Emiline (33)*, uracil (34), pedatisectine E (35), uridine (36). Among them,1and33were new compounds. All compounds were separated from this genus for the first time, except compounds14,23and29. Besides, pharmacological investigations showed that compound33at10.0μg·mL-1displayed noteworthy neuroprotective effect against the corticosterone-induced apoptosis in PC12cells.2. Online analysis on the common chemical constituents of E. sonchifolia and Huahong capsule was founded by UPLC/Q-TOF-MSE method. Four common caffeoylquinic acid derivatives in E. sonchifolia and Huahong capsule were confirmed by using elementary composition, MS/MS and relevant literatures. The results laid the foundation for quality control and further developing of herbal materials and preparations.3. The HPLC fingerprint of E. sonchifolia based on a well determination condition was established. The similarity of10batches of E. sonchifolia from different producing areas was set up by the software of "traditional medicine fingerprint similarity evaluation".18peaks were identified as the characteristic fingerprints of the10batches of E. sonchifolia, and the similarity was from0.851to0.992.4. An HPLC method for the simultaneous determination of three caffeoylquinic acid derivatives (neochlorogenic acid, chlorogenic acid and cryptochlorogenin acid) in E. sonchifolia and relevant preparations was built. The method was simple and sensitive, which was applied to determination the contents of three caffeoylquinic acid derivatives in E. sonchifolia and its preparations,10batches of each.5. The research progress on chemical constituents and pharmacological activities of plants belonging to genus Emilia were reviewed, providing useful references for the further research.
