Objective: To detect the expression of miR-27a and miR-146a in synovial tissue of the knee joint ofpeople with osteoarthritis (OA) and people with no osteoarthritis (OA),and investigate the correlationbetween the expression of them and the disease.Methods: Forty-six cases synovial tissue samples were collected from hospitalized patients ofDepartment of orthopedics the First Affilated Hospital of shihezi University from November2011to Mareh2012(Including23samples with osteoarthritis,23samples with no osteoarthritis), the expression ofmiR-27a and miR-146a in synovial tissue from46persons was measured by quantitative reversetranscription-polymerase chain reaction (RT-PCR). And to analyze the relationship between the severity ofosteoarthritis and the expression levels of miR-27a and miR-146a in synovial tissue. All the data were inthe form of”mean±standard”and analyzed with Independent sample t-test through SPSS17.0softwar. thePvalue less than0.05was considered statically significant.Result:(1) MiR-27a and MiR-146a can be detected in all knee synovial tissue. we found that theamplification curve show good and the amplification efficiency is higher.(2)The expression levels ofmiR-146a and miR-27a in OA synovial tissue is1.91±1.74、1.16±1.93,and in control group is0.91±1.19、0.13±1.35, according to theory of qRT-PC, the higher value of ΔCt was the lower expression of target genein corresponding tissue,which was that the expression of miR-27a and miR-146a in osteoarthritis patientssynovium of knee joint,compared with that of non-osteoarthritis was low trend(P<0.05).Conclusion: compared with non-osteoarthritis, the expression of miR-27a and miR-146a inosteoarthritis patients synovium of knee joint was low trend. The lower expression of miR-27a andmiR-146a in synovial tissue may be play a crucial role in pathologic changes of OA.
