Objective: To detect the expression levels of β-catenin and u PA in joint synovial tissue in patients with osteoarthritis(OA) and normal control group. Investigate the relationship of β-catenin and u PA with OA Patients synovial tissue lesions.Methods: OA group: articular synovial tissue were obtained from patients undergoing total knee arthroplasty(64cases). Control group: the normal knee joint cartilage, synovial tissue were obtained from patients undergoing trauma after amputation and femoral condyle fracture patients voluntary contributions in shihezi university school of medicine affiliated hospital(15cases).The articular synovial tissues were fixed and embedded in Paraffin. The sections were stained with Hematoxylin and Eosin(HE). The samples of articular cartilage were divided into three groups by using the modified Mankin score. There were control group, the mild OA group, middle and severe OA group.The expression levels of β-catenin and u PA were measured by immunohistochemistry and ELISA technology in synovial joints.The data was used X2 test and t test for statistical analysis.Result: 1. Articular cartilage destruction was found in OA patients, complete 1oss of articular cartilage, disordered arrangement of cells and gathered the clusters cell cloning, or tidemark breaking found in these patients. 2.The result of β-catenin immunohistochemistry showed negative in control group, positive in mild OA(the Positive rate 31.2%), 7 positive in moderate OA group(the Positive rate 77.8%) and positive in severe OA group(the Positive rate 87.5%%).The result of u PA immunohistoehemistry showed two positive in control group(the Positive rate1.7%), positive in mild OA(the Positive rate 31.2%), 21 positive in moderate OA group(the Positive rate 81.2%) and 7 positive in severe OA group(the Positive rate81.4%). 3.The synovial membrane content of β-catenin and u PA increased as well as the joint degeneration in patients with osteoarthritis, they were obvious positive correlation(P < 0.01).Conclusion: 1. The expression levels of β-catenin and u PA were increased in joint synovial tissue in patients with osteoarthritis(OA). The content of β-catenin and u PA were significant positive correlation with the lesion degree of OA, they worked together in the process of the cartilage and synovium degeneration in patients with osteoarthritis. 2.These results establish a strong association between Wni/β-catenin signaling and human OA.
