Objective:To observe the expression level of interleukin 17(IL-17), interleukin 19(IL-19) and insulin-like growth factor 1(IGF-1) of the osteoarthritic synovial tissue with lesions in different degrees, and to analyze the correlation between one of the above-mentioned factors and the bone mineral density(BMD), discussing the mechanism of action of those factors in osteoarthritis as well.Methods:60 cases of inpatients with osteoarthritis in joint surgery of maternal and child health hospital of Linzi district were selected as experimental group, and divided into 4 groups by Kellgren-Lawrence degree?,?, ? and ?. The synovial tissues of every patient were reserved in the surgery of knee artificial replacement. At the same time, 20 cases of patients with meniscus injury or ligamentous injury were selected as control group. The synovial tissues of every patient were reserved in arthroscopic surgery. The expression rate and levels of IL-17, IL-19 and IGF-1 were detected with immunohistochemical method. Further more, BMD of patients was detected before surgery, and the correlation between IL-17/IL-19 /IGF-1 or BMD were analyzed.Results:(1)BMD of patients in experimental group was lower than that in control group, and it decreased as the aggravation of illness, the difference between groups showed statistical significance(F=5.062, P<0.05).(2)The expression rate of IL-17 increased as the aggravation of illness, and was higher in experimental group than that in control group. There were significance between every double groups(x2=25.95/18.64, P<0.001). The expression rate of IL-19( x2= 13.075/14.468,P<0.05) and IGF-1(x2=19.588/17.357,P<0.05) decreased as the aggravation of illness, and was lower in experimental group than that in control group. There were significance between every double groups(x2=25.95/18.64, P<0.001). There were no significance between man and woman.(3) The expression levels of IL-17 increased as the aggravation of illness and was higher in experimental group than that in control group. There were significance between every double groups(F=22.145/16.354,P<0.001). The expression levels of IL-19 and IGF-1 decreased as the aggravation of illness, and was lower in experimental group than that in control group, There were significance between every double groups(FIL-19=22.145/18.425,PIL-19<0.001;FIGF-1=18.504/19.250,PIGF-1<0.001). There were no significance between man and woman.(4) The IL-17 expression level has negative correlation with BMD(R2 = 0.6778). IL-19 and IGF-1 expression level have positive correlation with BMD(R2 = 0.6503,0.6563).Conclusion:There is inflammatory response in the synovial tissues of patients with osteoarthritis. The degree of osteoarthritis pathological changes has negative correlation with BMD.
