Osteoarthritis OA is common and a kind of chronic disease which endangers elderly people. Pathologic changes of osteoarthristis of the knee mainly is the denaturation and distruction caused by the metabolic dysfunction of cartilage. There is no totally effective treatment for osteoarthristis OA by now. It is treated nunsurgically at the early stage, after reaching an advanced stage. The operative procedure was performed until cartilage is severely destroyed when it reaches an advanced stage, however, recently the treatment of diseases in bone and joint disease becomes a hot research spot worldwide. During lymphocytes proliferate and lymphoid differentiation Interleukin-7(IL18) plays a key role. B and T lymphocyte generate obstacles is caused by the surface of the cells lack of interleukin receptor, while severe combined immunodeficiency disease is because of lack of T cell, and the lack of T cell is mainly due to the defects in IL-7C。or C And studies have proved that the distinction lies in memory T cells and regulatory T cells, interleukin receptor also plays a role, the researchers are paying more attention to interleukin-7receptor mainly because it have the function of the differentiation of lymphocytes and the role of the body’s immune memory。 At present home and abroad articles about similar literature are rare. Therefore, we use gene chip, the organization haematin-eosin staining method, immunohistochemistry and RT-PCR method detectioon of IL-7R gene expression in the Osteoarthritis of knee joint synovial[Objective]Discuss osteoarthritis of knee joint synovial IL-R genes in knee osteoarthritis expression effect and abnormal expression in the process of occurrence and development.[Method]1. Collect64screening from Yanbian University Hospital orthopaedic patients fresh synovial tissue, and25cases of meniscus and cruciate ligament injury damage line arthroscopic surgery in patients with normal fresh synovial tissue,39cases reached OA diagnosis standard, the patients’synovium who underwent a line of knee artificial joint replacement, put in sterile tube after tissue in vitro, directly put in liquid nitrogen to-80℃refrigerator after30minutes.2. Store3cases of the synovial tissue of normal fresh synovial tissues and10cases of OA synovial tissue in liquid nitrogen to Beijing BO’AO LTD to detect gene chip.3. to confirmed gene chip results, perform Real time RT-PCR detection.4. knee joint synovial tissue must be put in four percent paraformaldehyde solution, and fixed in four degrees celsius temperature for24hours and dehydrate and embedded in paraffin. Finally, slice all the specimensnto sections using microtome according to4um of specifications and fixed on the slide.5. Fixed on the slide and used in the HE staining and immunohistochemical experiments. Special attention should be paid to that the used biopsy must be put in60degrees ceisius temperature box。6. Use trizol method to extract total RNA in the synovial tissue. Use the applied biosystems retrovirus kit retrovirus the first chain of Cdna. Applied by PCR amplification reaction to increase IL-7R genes, and reverse transcription polymerase chain reaction. Detect54cases of knee patellar capsule in synovial tissue IL-7R expression and the significance of IL-7R genes in knee joint osteoarthritis。.[Result]1. It can be seen in mRNA gene chip detection results that compared with synovial tissue in normal knee joint, its expression declined obviously in osteoarthritis knee joint。2. Gene expression in knee OA synovial tissues decreased obviously。3. HE staining results show that inflamed and degenerated synovial tissue, synovial cell morphology changed and inflammatory cells infiltrated associated with connective tissue hyperplasia and hypertrophy.4. Immune physiochemical method results show that IL-7R expression in synovial cell cytoplasm after osteoarthritis knee joint inflammates IL-7R expression decline.5. RT-PCR experiment results show that compared with normal knee joint synovial tissues, IL-7R in knee OA synovial membrane is significantly decreased.[Conclusion](1) During the whole process of knee osteoarthritis treatment, reactive hyperplasia is obvious in patients’synovial tissue。(2) This study found that IL-7R expression in the osteoarthritis of knee joint synovial cells declined obviously。(3) The IL-7R expression in the osteoarthritis of knee joint synovial tissue decreased obviously and it played an important role in knee osteoarthritis pathological process. Regard the IL-7R as one of the key points of early diagnosis and treatment of knee osteoarthritis disease, and provide a new way for the clinical treatment of knee osteoarthritis.
