Objective: To detect the expression significance of miR-27aand miR-146a in cartilage tissue of kneejoint of different stage for the normal and osteoarthritis(OA) model in SD rat,and study the effects ofmiR-27a and miR-146a in development and progression of osteoarthritis.Methods: The80SD rat were divided into experimental animals and control animals,60cases forexperimental group and20cases for control group. Experimental animals were randomly divided into threegroups acording to time, three experimental groups were transected of the medial collateral ligament,medial meniscectomy, anterior cruciate ligament and posterior cruciate ligament. The rats were killed at the4st,6nd and8th weeks postsurgery to obtain the right knee joints.the expression of miR-27a and miR-146ain cartilage tissue was measured by quantitative reverse transcription-polymerase chain reaction (RT-PCR).And to analyze the relationship between the severity of osteoarthritis and the expression levels of miR-27aand miR-146a in cartilage tissue. All the data were in the form of “mean±standard”through SPSS17.0softwar. the Pvalue less than0.05was considered statically significant.Result:(1) MiR-27a and MiR-146a can be detected in all knee cartilage tissue.(2)The expression level of miR-27a and miR-146a in experimental group is much more lower comparedwith control group(P<0.05).(3)The expression level of miR-27a,miR-146a and the severity of OA was negatively correlated.(r=-0.32,r=-0.67,P<0.05)Conclusion: The expression level of miR-27a and miR-146a in experimental group is lower thancontrol group and have great correlation with the severity and progression of osteoarthritis(OA),It may bean important indicator of early diagnosis and the activity of the disease of osteoarthritis.
