Objective:To investigate the expression of fibroblast growth factor-1(FGF-1) and Calcium binding protein S100A13(S100A13) in cholangiocarcinoma (CCA) and analyze the correlation between them,to explore the relationship in the development of cholangiocarcinoma, in order to provide basis for clinical prevention and treatment of cholangiocarcinomaMaterials and Methods:Collected surgical specimens from Hunan Provincial People’s Hospital during June2008-June2012.The experiments were divided into three groups:normal group (biliary bile duct trauma and iatrogenic bile duct injury specimens)(group A), corresponding paracarcinomatous Bile duct tissues (B group), cholangiocarcinoma(C group). S100A13mRNA and FGF-lmRNA in20fresh CCA specimens which consisted of cancer tissues and corresponding paracarcinomatous Bile duct tissues,and10Normal bile duct tissues, were detected by Real-time PCR(Polymerase chain reaction). The expression of S100A13protein and FGF-1protein were examined in40paraffin CCA specimens, which also consisted of cancer tissue and corresponding paracarcinomatous Bile duct tissues, and20normal Bile duct tissues by immunohistochemistry. And analyzing the results of the test, to observe the expression of S100A13FGF-1in bile duct carcinoma, and to analyze its correlation with clinicopathological characteristicsResult(1) The mRNA of S100A13and the mRNA of FGF-1in cholangiocarcinoma tissues were Significantly increased compared with paracarcinomatous Bile duct tissues and normal bile duct tissues,The Real-time PCR shows a strong Positive correlation between the expression of S100A13and that of FGF1in CCA by the Person correlation test analysis(r=0.89, P=0.045). In the A group, B group and C group, The expression of S100A13protein was significantly stronger in cancer tissues than in paracarcinomatous Bile duct tissues and normal bile duct tissues with the positive rate80%,30%and25%respectively, the differences were statistically significant (P<0.05). The expression of FGF-1protein was significantly higher in cancer tissues than inparacarcinomatous Bile duct tissues and normal bile duct tissues with the positive rate82.5%,30%and30%respectively, the differences had statistical significance (P<0.05).(2)The expressions of S100A13and FGF-1in cholangiocarcinoma were not relative to the gender,age, tumor pathological types and tumor differentiation degree(P>0.05),but their expressions in bile duct carcinoma were significantly correlated with lymph node metastasis and portal vein invasion (P<0.05)(3) The high expression of FGF1was followed by a high expression of S100A13in the tissues of cholangiocarcinoma.The immunohistochemical staining shows a strong Positive correlation between the expression of S100A13and that of FGF1in CCA by the Pearson correlation test analysis (r=0.63, P<0.01).Conclusion(1) S100A13protein and FGF-1protein were highly expressed in cholangiocarcinoma.(2) In cholangiocarcinoma, high expression of S100A13protein and FGF1protein are more susceptible to lymph node metastasis and vein invasion(3)There may be a positive correlation between the expression of S100A13and that of FGF-1in cholangiocarcinoma...
