Objectiveâ‘ To explore the expression and localization of hepatocellular carcinoma-associated antigen S100A13 in normal liver, HCC and their corresponding paracancerous tissues, so as to evaluate its potential ability for diagnosing and treating liver cancer.â‘¡To research the expression and distribution of TMEM135 mRNA and protein in a variety of normal and tumor tissues. Methodsâ‘ S100A13 mRNA and protein expression levels were detected in normal liver, HCC and their corresponding paracancerous tissues respectively by using the methods of in situ RT-PCR and immunohistochemistry.â‘¡The expression profile of TMEM135 mRNA and protein was analyzed in a variety of tissues by using the methods of in situ RT-PCR and immunohistochemistry respectively. ResultsThe positive rate of S100A13 mRNA was 70.21%, 51.06% and 33.33% respectively, in 47 cases of HCC and their corresponding paracancerous tissues, 9 cases of normal liver tissue. The positive rate of S100A13 protein was 55.84%, 38.96% and 27.78% respectively in 231 cases of HCC and their corresponding paracancerous tissues, 18 cases of normal liver tissue. By analyze the average optical density value above three tissues by Image-Pro Plus software,we found that the average optical density value in HCC tissue was the highest. The expression of S100A13 protein was the highest in HCC, followed by paraneoplastic tissue, and normal liver tissue was expressed the lowest. Furthermore, S100A13 protein also was highly expressed in the adjacent the secondary bile canaliculi and part of inflammatory cells. There was no significant difference between S100A13 expression in HCC and its tumor grader. S100A13 protein was mainly expressed in the cytoplasm and, occasionally expressed in some nucleus. TMEM135 mRNA and protein expressed were detected in 36 cases of normal tissues, 12 cases of benign tumor tissues and 34 cases of cancer tissues by the methods of in situ RT-PCR and immunohistochemistry, respectively .We could see that TMEM135 mRNA and protein expressed widely in human tissues except the pituitary gland, but the protein was appeared weak expression comparing to mRNA expresion. TMEM135 mRNA expression was mainly distributed in the nuclear membrane, and the protein was mainly distributed in the cytoplasm with many granular distribution. The positive rate of TMEM135 mRNA in HCC, paraneoplastic and normal liver was 45%, 30% and 50% respectively, and the positive rate of TMEM135 protein was 45%, 35% and 50% respectively. The expression of TMEM135 protein in tissues was associated with age. Conclusionsâ‘ S100A13 protein is associated with HCC, and it has the potential to serve as a target of cancer therapy.â‘¡TMEM135 is a new protein which has a great of value in scientific research and has potential value at diagnosis and therapy of cancer and aging research.
