| Objective:①to obtain CD147expression in the related data ofhepatolithiasis and intrahepatic cholangiocarcinoma;②Analysis of CD147and MMP-2, the relationship between MMP-9expression, a relationshipwithout metastasis, and CD147, MMP-9, MMP-2expression and patient age,gender, bile duct cancer tumor size, tumor differentiation,cholangiocarcinoma;③preliminary assessment of CD147in the diagnosisand treatment of intrahepatic bile duct stones with bile duct cellcarcinoma.Materials and methods: Collection of hepatolithiasis associated withcholangiocarcinoma operation resection specimens from30patients,collected from normal liver tissue specimens of10cases of operation(experimental samples were all from the operation room of Hunan People’sHospital operation resection specimens). Group: A group: hepatolithiasiswith bile duct cell carcinoma (n=30); group B: hepatolithiasis combinedwith cholangiocarcinoma cancer tissues (n=30, para cancer tissue fromtumor edge is about5cm region of the normal liver tissues, and allconfirmed by pathological examination revealed no cancer cells in Cgroup);: normal liver tissues (n=10). Collect and provide samples ofpatient age, gender, bile duct cancer tumor size, tumor differentiation,bile duct cancer cells have no clinical pathological data transfer. Threegroups of CD147mRNA were used to detect the expression of qRT-PCR wasdetected by immunofluorescence method, three group of CD147proteinexpression; Western-Blotting assay in three groups of matrixmetalloproteinase9(-2, MMP-2, MMP-9) protein expression. Analysis of CD147and MMP-2, the relationship between the expression of MMP-9and bileduct cell carcinoma, and the relationship between CD147, MMP-9, MMP-2expression and pathological data of patients.The research results:1.QRT-PCR detection showed, intrahepatic bile duct cell carcinoma CD147protein expression was higher in bile duct carcinoma paracanceroustissues and normal liver tissues, bile duct cancer adjacent tissues CD147expression was higher than that of normal liver tissues. CD147bile ductcell carcinoma and the tumor size, lymph metastasis and tumordifferentiation, tumor growth, lymph node metastasis, tumordifferentiation degree low expression of CD147in cholangiocarcinoma.CD147bile duct cell carcinoma was not related to age and sex.2. Immunofluorescence detection showed intrahepatic bile duct cellcarcinoma CD147protein expression was higher in bile duct carcinomaparacancerous tissues and normal liver tissues, bile duct cancer adjacenttissues CD147expression was higher than that of normal liver tissues.3.Detection of Western blot showed that the bile duct cancer cells ofMMP2and MMP9protein expression was higher in bile duct carcinomaparacancerous tissues and normal liver tissues, bile duct carcinomaparacancerous tissue MMP2and MMP9expression was higher than that ofnormal liver tissues.4. Through the statistical analysis of correlation was positivelycorrelated with the expression of CD147and MMP2, suggesting that MMP9in the same cell carcinoma of bile duct.Conclusion:1. Enhanced expression of CD147, MMP2, MMP9in intrahepatic bile duct cellcarcinoma, the expression of CD147may promote the up regulation of MMP2,MMP9expression.2. CD147may increase as the patient tumor, lymph node metastasis, and increased expression of the low degree of tumor differentiation. |
