Objective:To explore the expression of mitogen activated protein kinase kinase5(MEK-5), extracellular signal-regulated kinases5(ERK5),phospho-extracellularsignal-regulated kinase(p-ERK), vascular endothelial growth factor (VEGF),and matrix metalloproteinases9(MMP-9) in eutopic and ectopic endometrium of endometriosis and the relationship between them.Methods:The protein level of MEK-5, ERK5,p-ERK,VEGFand MMP-9were detected by immunohistochemical SP method in formalin-fixed,paraffin-embedded samples of40eutopic and ectopic endometrium of EMs,40normal endometrium. Combined with clinical cases,computer pathological image analysis system was used in this study.All data were analysed by statistic software SPSS, version17.0.Results:1. MEK-5, ERK5,p-ERK,VEGF and MMP-9were detected in three groups, and they were mainly observed in glandular epithelial cells,mostly localized in cytoplasm.2.The expression of MEK-5, ERK5, p-ERK and MMP-9in EMs ectopic was significantly higher than the eutopic endometrium(P<0.01), the eutopic endometrium was significantly higher than the normal endometrium(P<0.01). There was no significant difference of VEGF expression between the eutopic and ectopic endometrium,but they are both higher than normal endometrium(P<0.01).3. The expression of MEK-5,ERK5,p-ERK and VEGF were positively related,also positive correlation between the expression of MEK-5,ERK5,p-ERK and MMP-9. The expression of VEGF and MMP-9also had a synergy correlation in the three groups of endometrium.Conclusions:1. The expression of MEK-5、ERK5and p-ERK in EMs eutopic and ectopic endometrium were significantly higher than the control group, suggested that there was certain correlation between the ERK5signaling pathway and EMs.2. VEGF and MMP-9were high expressed in ectopic and eutopic endometrium. And there were positive correlation between the expression of VEGF,MMP-9and MEK-5,ERK5,p-ERK. It suggested that there were mutual effection between the ERK5signaling pathway and VEGF、MMP-9. They all participated in the occurrence and development of EMs.
