Objective:To detect the expression of angiopoietin-2 (Ang-2),vascular endothelial growth factor(VEGF)in endometriosis and investigate both the relationships of angiogenesis in endometriosis.In order to know better the pathogenesy of endometriosis,and provid theory to clinical diagnosis and therapy.Methods:Elivision Tm plus in Imunohistychemistry method and Stereology method was used to detect the expression of Ang-2,VEGF and CD34 in 30 cases of ectopic endometrium,30 cases of eutopic endometrium and 30 cases of endometrium in the normal control.MVD was assayed by CD34 in positive blood vessel.Quantitative images analytical system was performed with computer to analysis the mean optical density of Ang-2,VEGF and MVD in Stereology.Results:(1)Ang-2 was found predominately in the cell-substance of interstitial cell,epithelioglandular celland blood vessel endothelium cell.It can be found primary in the interstitial cell.The expression of Ang-2 in ectopic endometrium of endometriosis is obviously higher than the normal control.The expression of Ang-2 is (0.32±0.08),(0.47±0.12) and (0.39±0.14) which in ectopic endometrium of endometriosis,eutopic endometrium of endometriosis and the normal control .The expression of Ang-2 in secretory phase of eutopic endometrium of endometriosis and the normal control are higher than which in the proliferative phase,the difference has statistical significance(P<0.05).The eutopic endometrium of women with endometriosis both in the proliferative phase and secretive phase expressed higher level of Ang-2 than that of the control gorup(P<0.05).(2)VEGF was found predominately in the endochylema of glandular epithelium and vascular endothelial cell.It also can be found in the mesenchymal cell of surrounding.VEGF in the glandular epithelium is more than that in the mesenchymal cell.The expression of VEGF is (0.32±0.08),(0.47±0.12 ) and (0.39±0.14) which in ectopic endometrium of endometriosis,eutopic endometrium of endometriosis and the normal control .The expression of VEGF in eutopic endometrium of endometriosis and in control group was higher than that of ectopic endometrium of endometriosis﹙P<0.01 or P<0.05﹚.The expression of VEGF in secretory phase of eutopic endometrium of endometriosis and the normal control are higher than which in the proliferative phase,the difference has statistical significance(P<0.01).The eutopic endometrium of women with endometriosis both in the proliferative phase and secretive phase expressed higher level of VEGF than that of the control gorup(P<0.05). (3)CD34 can be foud in the cell membrane and intracytoplasm of various kinds of vascular endothelial cells. The expression of MVD is (41.58±14.72),(31.30±19.89)and(20.70±16.17),which in ectopic endometrium of endometriosis,eutopic endometrium of endometriosis and the normal control(P<0.01).The expression of MVD in secretory phase is higher than which in proliferative stage of each group(P<0.01).The eutopic endometrium of women with endometriosis both in the proliferative phase and secretive phase expressed higher level of MVD than that of the control gorup(P<0.01)(.4)Relative analysis revealed that:The correlation coefficient of ang-2 and VEGF is 0.629(P<0.05),0.944(P<0.01),0.257(P>0.05)in ectopic endometrium of endometriosis,eutopic endometrium of endometriosis and the normal control;The correlation coefficient of ang-2 and MVD is 0.945(P<0.01),0.648(P<0.05),0.376(P>0.05)in ectopic endometrium of endometriosis,eutopic endometrium of endometriosis and the normal control;The correlation coefficient of VEGF and MVD is 0.66(P<0.05),0.875(P<0.01),0.561(P<0.05)in ectopic endometrium of endometriosis,eutopic endometrium of endometriosis and the normal control.Conclusion:(1)Ang-2 was found predominately in the cell-substance of interstitial,celepithelioglandular celll and the blood vessel endothelium cell.It can be found primary in the interstitial cell.The expression of Ang-2 in ectopic and eutopic endometrium group was higher than which in the control group,Ang-2 has a close correlation with the angiogenesis of women with endometriosis.(2)VEGF was found predominately in the endochylema of glandular epithelium and vascular endothelial cell,It also can be found in the mesenchymal cell of surrounding,In the glandular epithelium the expression of VEGF is more than that in the mesenchymal cell.VEGF has a close correlation with the angiogenesis of women with endometrio -sis.The expression of VEGF in the eutopic endometrium of endometriosis is the highest of all.The eutopic endometrium of women with endometriosis were different from the control group,and the special quality of the endomembrane maybe the take a essential condition in the development of endometriosis.(3)The expression of MVD in the ectopic and eutopic endometrium is much higher than which in the control group.It suggests that there is a prerequisite of new vascular formation in the course of the angiogenesis in endometriosis. (4)The expression of Ang-2 has a direct correlation wth the expression of VEGF and MVD,which means that there maybe has a connection between Ang-2 and VEGF, they may participated the angiogenesis of endometriosis,and they could promote the development of endometriosis together.
