Objective: To investigate the antipsoriatic activity of compoud glycyrrhizin(SNMC), and further study its mechanism.Methods: A facility was devised for culturing the psoriatic skins. According topsoriatic pathogenesis, psoriatic lesions were cultured at the air/liquid interface inserum-free William’s medium supplemented with psoriatic T cells and streptococcalantigens. After the psoriatic skins model was successed, co-incubate with SNMC indifferent concentrations(100,200and500ml/L) for6days, and one group withoutSNMC as control. After incubation, psoriatic characterization was analyzed bymacroscopic observation and light microscope. Furthermore, detected keratin17expressed in psoriatic skin by immunohistochemistry method to examine the effect ofSNMC on antipsoriatic. Meanwhile, the contents of cytokine interleukin-10andinterleukin-17were detected by enzyme-linked immunosorbent assay (ELISA).Analyzed all the data by SPSS and EXCEL statistically.Results: From the result of macroscopic observation, the psoriatic lesions incontrol group is smooth and shiny, but groups co-incubation with SNMC were wizenedand gloomy. Psoriatic lesions which cultured at the air/liquid interface in serum-freeWilliam’s medium supplemented with psoriatic T cells and streptococcal antigens after6days still maintained the psoriatic structure and characterization observed by lightmicroscope, but SNMC can inhibit these symptom. The results ofimmunohistochemistry method showed that high expression of K17is noted in thecultured psoriatic lesions, and there is significant difference with SNMC500andSNMC200(p<0.01). The results of enzyme-linked immunosorbent assay showed thatSNMC can decrease the expression of IL-17(59.82±6.73vs219.82±9.56pg/ml),while increase the content of IL-10(16.21±0.72vs8.41±0.80pg/ml).Conclusion:1The study validate the antipsoriatic effect of SNMC by the vitro psoriatic organ culture model.2SNMC can decrease the expression of K17and IL-17in psoriatic lesions,meanwhile increase the content of IL-10. The result showed that the antipsoriaticmechanism of SNMC might be closed linke to regulate these cytokines.
