Compoud Chrysanthemum indicum soft capsule (CISC) is a Chinese patent medicine consisting of the CO2-supercritical fluid extract of Chrysanthemum indicum (SFEC), Patchouli oil (PO) and Zedoary Turmeric oil (ZTO). According to the previous studies, it has been proved that SFEC and PO have the excellent anti influenza viral pneumonia activities while CISC owned a more outstanding effect. Based on the above findings, the overall pharmacodynamics of CISC under the guide of traditional Chinese medicine was performed and the results were as follows.1. Influenza symtoms remissionFirstly, CISC could significantly inhibit the pulmonary pathological changes and decrease the death rate caused by influenza A virus. However, CISC had no suppression on hemagglutination caused by influenza A virus under its maximum non-toxic concertration of 1.56μg/mL on MDCK.Secondly, CISC could also protect the mice from the death caused by staphylococcus aureus infection and hemolytic streptococcus-βinfection by means of decreasing the death rates and expanding the longevity. The minimal inhibitory concentrations (MIC) on hemolytic streptococcus-βand streptococcus pneumoniae were 2.5 mg/mL respectively and the MICs of staphylococcus aureus and Candida albicans were 20mg/mL, while CISC was observed no inhibiton on colibacillus, bacillus pyocyaneus and micrococcus catarrhalis. Thirdly, CISC attenuated the fever coused by the exogenous pyrogen (yeast on rats and lipopolysaccharide on rabits) suggesting that it could prevent the secondary fever after infection.2. Immunity regulation and anti-inflammationIn the aspect of immunity regulation, CISC could not only restore the phagotrophy function of macrophage in serum proved represented by the elevating expurgation index, but also lift the serum concentration of IgG and IgM. On the second place, CISC could restrain the ear hemangiectasis induced by DNCB while no inhibiton of exudation after histamine subcutaneous injection challenge was observed, data from which suggested that CISC had a supression on the delayed type hypersensitivity (DTH) but not on immediate hypersensitivity. What’s more, CISC could not only decrease the dimethylbenzene (DMB)-induced ear vasodilatation but also attenuat acetic acid-induced capillary permeability in mice. And an obvious inhibition on carrageenan-induced paw edema was observed. ELISA assays showed that CISC could up-regulate the production of IL-1βsignificantly while down-regulate that of PGE2 in the edematous paw tissue, but no apparent effect on TNF-a or NO was observed in the test. Besides, CISC could suppress the granuloma formation induced by cotton pellet.3. Relieving cough and reducing sputumCISC could prolongate the cough latent time and reduce the cough times after NH3 and SO2 challenges. On the other hand, CISC could increase the phenol sulfonphthalein exudation in the trachea of mice and the travelling distance of carbonparticle in the trachea of pigeons.All results above indicated CISC had a potent anti-inflenza activity as well as the medical properties on against secondary bacterial infection and fever. Besides, CISC had promotion effects both on the specific and non-specific immunity at the same time an apparent anti-inflammatory activity. Lastly, results of these experiments also suggested that CISC was an excellent antitussive and expectorant.In conclusion, the anti-influenza activity of CISC was based on the properties of anti-virus pneumonia, anti-bacteria infection, as well as fever attenuation, supporting by cough relieving and phlegm eliminating. And the key point of its anti-influenza activity may be laid on the immunity regulation and inflammation lesion remission.
