Objective: To study the expression and correlation of ALDH1、HIF-1α、ABCG2and VHL in breast stem cell and their affection in the mechanisms of drug resistance;Methods: Immunohistochemistry (SP method) were used to detected the stem cellmarker ALDH1, resistance-associated protein of ABCG2, HIF-1α and VHL in15casesof normal breast tissue,30cases of mammary hyperplasia and40cases of breastinvasive ductal carcinoma, the expression and correlation of ALDH1, ABCG2, HIF-1αand VHL in breast tissue were analyzed. Explore the breast cancer stem cells and therelevance of the multidrug resistance escape mechanism.Results:1、The rate of ALDH1positive expression in normal tissues was6.7%,hyperplasia was30.0%, cancer tissue was57.5%, the expression of ALDH1in normaltissue, hyperplastic tissue, cancer tissue has statistical difference(χ~2=13.389,P=0.001),the rate of ALDH1positive expression in invasive ductal carcinoma tissue is rising withpathological level, the expression in different pathological grading organization has astatistical difference(χ~2=8.995,P=0.011).2、The rate of HIF-1α positive expression in normal tissue, hyperplasia, and cancertissue was6.7%,26.7%,57.5%. the difference between positive expression rate werestatistically significant(χ~2=14.390,P=0.001). HIF-1α positive expression in invasiveductal carcinoma tissue is rising with pathological level, the difference between datawere statistically significance(χ~2=6.227,P=0.038).3、The rate of ABCG2positive expression in normal tissue, hyperplasia, andcancer tissue was13.3%,40.0%,55.0%.The expression of ABCG2in hyperplasia andinvasive is higher than normal tissue, the difference between data were statisticallysignificance (χ~2=7.862,P=0.020). the positive expression of ABCG2in pathologicalgrading for grade II, III was significantly higher than that of grade I, the difference between data were not statistically significance(χ~2=0.684,P=0.710).4、The rate of VHL positive expression in normal tissue, hyperplasia, and cancertissue was93.3%,80.0%,52.5%,the difference between positive expression rate werestatistically significant(χ~2=8.547,P=0.014). VHL positive expression in invasiveductal carcinoma tissue is decreasing with pathological level, the difference betweendata were statistically significance(χ~2=5.830,P=0.043).5、The rate of ABCG2positive expression is positively correlated with ALDH1positive expression in pathological grade, clinical stage, tumor size, lymph nodemetastasis. The rate of ABCG2positive expression in the ALDH1-negative breastcancer cells was17.6%, in the ALDH1-positive expression rate was82.6%, thedifference between data were statistically significance (P=0.000).6、The rate of ABCG2positive expression is positively correlated with HIF-1αpositive expression in pathological grade, clinical stage, tumor size, lymph nodemetastasis. The rate of ABCG2positive expression in the HIF-1α negative breast cancercells was11.8%, in the HIF-1α-positive breast cancer cells was87.0%, the differencebetween the data was statistically significant (P=0.000).7、The rate of VHL positive expression is negatively correlated with HIF-1αpositive expression in pathological grade, clinical stage, tumor size, lymph nodemetastasis. The rate of VHL positive expression in the HIF-1α negative breast cancercells was100.0%, in the HIF-1α-positive breast cancer cells was17.3%, the differencebetween data was statistically significant (P=0.000).
