Objective: To investigate the relationship between ALDH1highCD44+CD24-/low breast cancerstem cells from pleural fluids or peripheral blood of breast cancer patients and clinicalsignificance.Methods:(1)sample①.single cells of MCF-7 cell line.②.collect 58 blood samples from 12volunteers and 46 breast cancer patients.③.collect 10 pleural fluids samples from breastcancer patients.(2)method The human MCF-7 cells is used as a model to develop a suspension arrayassay for the detection of markers expression for human breast cancer stem cells ,then oursuspension array is used to examine the markers expression for the cells from peripheralblood or pleural fluids of breast cancer patients.(3)content investigate the relationship between ALDH1highCD44+CD24-/low cells fromperipheral blood or pleural fluids of breast cancer patients and clinical significance,diagnosisvalue.Results:(1) The expressions of ALDH1highcells in the direct analysis group after ALDH1 reactionand the further labeled antibody group were(67.5±1.3)%and(66.1±1.5)%,respectively.There is no significant difference between 2 groups(P>0.05);The expressionof CD44+CD24-/lowwere(9.1±1.6)% in the direct labeling antibody group and(8.9±1.1)%in the ALDH1+antibody group,respectively.There is no significant difference between 2groups(P>0.05) .(2) The proportion of ALDH1highCD44+CD24-/low cells of volunteers ,Ⅰphrase,Ⅱphrase,Ⅲphrase,Ⅳphrase breast cancer patients are 0.00%,0.00%0.04%,0.00%0.13%, 0.00%1.45%,0.19%6.80%,respectively; The content of ALDH1highCD44+CD24-/low cellshave statistical significance betweenⅣphrase breast cancer patients'with all otherteams(P<0.05); All other teams are no statistical significance among them(P>0.05).(3)Among the ten cases of breast cancer patients'pleural fluids,the proportion ofALDH1highCD44+CD24-/low cells is between 0.14% to 6.80%,and the mean average is 2.56%.There is no statistical significance between ALDH1highCD44+CD24-/low cells proportion withER or HER-2 status(P>0.05).Conclusions:(1) We successfully develop a suspension assay for the detection of markers expression forhuman breast cancer stem cells.The detection results of ALDH1highCD44+CD24-/lowcells could not be impacted by further labeling antibody after ALDH1 reaction,and thedetection results of further labeled antibody also could not be impacted by ALDH1reaction.(2) The 11 samples of theⅣphrase breast cancer patients'peripheral blood are all detectedthe aim cells,and 4 of them form groups in the FCM pictures.The detection ofALDH1highCD44+CD24-/low cells in breast cancer patients'peripheral blood might beuseful tool for diagnosising and evaluating the curative effect.(3) ALDH1highCD44+CD24-/low cells might be the common breast cancer stem cells of all thebreast cancer gene subtypes.
