A Comparison Between Raloxifene And Clomiphene Citrate Forovulation Induction In Polycystic Ovary Syndrome

Objective: To compare the ovulation rate between raloxifene(RAL) andclomiphene citrate (CC) in patients with polycystic ovary syndrome(PCOS).Methods: The patients with PCOS undergoing ovarian stimulation wererandomized to treatment with RAL120mg/d(n=31),CC50mg/d (n=32) for5days starting on day3of menses.The following indexes were compared:thenumber of follicle≥14mm and follicle≥16mm、thickness of endometriumwere observed and record on the day of HCG、the concentration of serumP on day7after follicular rupture. Meanwhile, pregnancy rate wererecorded as well as the case of miscarriage and ovarian hyperstimulationsyndrome(OHSS).Results: The women with PCOS were randomized to receive RAL(33cycles)orCC(41cycles).From these,65cycles finished the trial(RAL: n=29;CC:n=36).The ovulation rates of group RAL（27.3%）is lower than group CC（58.5%）,according to an intention-to-treat analysis,with statisticallysignificant differences between the groups.While the per-protocolrustles were different,the ovulation rates of group RAL is also lower thangroup CC,but there were no statistically significant differences betweenthe groups in ovulation rates per-protocol(RAL31%,CC52,8%).Comparedwith the CC group, RAL was associated with a statistically significantlyfewer follicles≥14mm. The endometrial thickness of CC group is thickerthan group RAL at day HCG,and the difference has statistical significant.The pregnancy rate and ongoing pregnancy rate were lower in the RAL group,but this has no statistical significance. No one patient with OHSS in RALgroup,while1occurred in CC group.No serious adverse events were observedin either group.Conclusion: RAL results in stimulation of ovarian folliculogenesis. RALwas associated with a statistically significantly lower ovulation rates than CC,but also associated with a less influence in endometrium, Theendometrial thickness of CC group is thicker than group RAL at day HCG,andthe difference has statistical significant.The pregnancy rate were lowerin the RAL group, but this has no statistical significance.The suitabledosage of RAL for ovulation induction need to confirmed.
