Background: Cerebrovascular disease (also called stroke) is the high rate of morbidity,and67.3%-80.5%of the ischemic stroke. Atherosclerosis is considered to be pathologyfoundation of ischemic cerebrovascular disease, its pathogenic mechanism is vascular stenosisand plaque off the embolism.The function of Matrix metalloproteinase (MMPs) is degrading andshaping the extracellular matrix (ECM) and maintain the dynamic balance of the extracellularmatrix. MMPs expression increased, which can make the fibre composition of degradation,promote the artery atheromatous plaque formation, and cause it to burst and the formation ofthrombus. Shedding of blood clots which can lead to cerebral infarction and other related disease.In the MMPs family,MMP-9expression is the most widely, higher than other types ofMMPs.To seek more prediction method,this research which take MMP-9gene polymorphism asthe base to study the relationship of unstable plaque and formation of micro embolus, furtherexplore the pathogenesis of carotid atherosclerosis especially unstable plaque,and geneticcharacteristics of micro embolus.Objective:To investigate the relationship between polymorphism at promoter ofC1562T of Matrix metalloproteinases-9(MMP-9) with positive of Carotid atherosclerosismicro embolus to provide the help for cerebrovascular disease prevention, early diagnosis andtreatment.Methods:A diagnosis of carotid ultrasound ischemic cerebrovascular were done fordisease patients, and they were screened with120patients who have unstable plaques withcarotid atherosclerosis. Further, color transcranial doppler ultrasound (TCD)were used tomonitor micro embolus.Dividing63examples of the positive of micro embolus patientsgroup and57of the negative of micro embolus patients group. At the same time application polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP)technology was applied to type gene sites of MMP-9polymorphism,and extract10%therandom samples to confirme genotype of promoter C1562T of MMP-9.Finally, using thestatistics to compare the two groups genotype and allele frequency, and analyzes the promoterC1562T of MMP-9the polymorphism of whether there is a correlation with micro emboluspositive.Results: Micro embolus positive group whose CT type and TT types significantly higherthan the control group. Two groups have statistical significance(χ2=32.568P=0.000,P<0.05). The T allele of MMP-9is a risk factor of the formation of micro embolus(OR=10.000,P<0.05).Conclusion: MMP-9gene promoter C1562T sites intimately related to the formationof micro embolus.
