Cotyledon and hypocotyl of perilla as explants materials, the optimal conditions forcallus induction of perilla were determined by using plant tissue culture techniques inthis research, and the nutrients composition and active ingredient of hypocotyls inducedcallus of10perilla varieties. The results were obtained as follows:(1) MS medium was suitable for the callus induction of perilla hypocotyl andcotyledon, and the best callus growth conditions were12h/d lighting, pH5.8and25±1℃. The callus induction ability of hypocotyls was higher than that of cotyledons.The best hormone combination of hypocotyl and cotyledon callus induction were6-BA3.0mg/L+NAA0.3mg/L and6-BA2.0mg/L+2,4-D1.0mg/L, and in these conditionsthe induction rate was up to100%. ZB-1existed the best ability to inducing hypocotylscallus among10different perilla genotypes.(2) The callus induced from perilla hypocotyl were subcultured and the resultsshowed that the increasing amount of callus was improved with the increasement ofinoculation density. That was the largest when the inoculation density was1.2g/dish.The best time for callus harvest was the24th day, then the callus growing well and hadloose texture.(3) There was a highly significant difference all nutrients contents among10perilla varieties callus, fat ranged from4.44%~7.56%, cellulose12.60%~19.46%,protein0.433%~0.600%, polysaccharide16.92%~20.73%and VC21.15mg/100g~40.67mg/100g, respectively. The fat, cellulose and protein content of ZB-2were the highest among10varieties.(4) There was a highly significant difference in active ingredients among10perilla varieties callus, total flavonoid ranged from12.22mg/g~64.05mg/g,polyphenol15.77mg/g~73.73mg/g, proanthocyanidin1.12mg/g~1.86mg/g, rosemary acid1.81mg/g~10.54mg/g and sterol1.70mg/g~6.68mg/g. In addition toproanthocyanidin, the other4active ingredients of ZB-1callus were the highest among10perilla varieties, so ZB-1callus could be the best material for the perilla activeingredients study.
