Spatholobus suberectus Dunn has efficiency of invigorating the circulation of blood,which can increase the red blood cells and hemoglobin. The main role of Spatholobussuberectus Dunn is flavonoid compounds, which has effects of anti-tumor, anti-virus,anti-oxidant, curing cadiovasular disease and lower cholesterol. Spatholobus suberectusDunn are as raw materias to detect the conditions of flavonoids in Spatholobus suberectusDunn by Sodium nitrite-aluminum nitrate colorimetric method. Meanwhile, theextraction and separation process conditions, and the antioxidant conditions in differentextraction parts of Spatholobus suberectus Dunn also studied in the paper. The results arebelow:1. Selection of detecting the conditions of flavonoids in Spatholobus suberectus Dunnby Sodium nitrite-aluminum nitrate colorimetric method. Research Result Shows that themaximum absorption of complex of aluminium nitrate and flavonoids was at510nm, andthe Colorimetric condition were:5%sodium nitrite,10%aluminum nitrate,1mol/L NaOH,15min Chromogenic time and50%alcohol to meter volume. When detected the totalcontent of flavonoids in these conditions, the accuracy, repeatability and the RSD ofrecovery are0.92%,0.48%and5.0%,respectively. The stability of results are high2. Selection of extraction solvent of flavonoids in Spatholobus suberectus Dunn.When the ration of material was1:30, the temperature was50℃,the extraction time was120min, the50%alcohol is the best impregnation method among water,95%alcohol,50%alcohol, acetone, chloroform, ethyl acelate,diethyl ether, petroleum ether eight solventsby compareing the extracton raton of flavonoids.3. Optimization of extraction technology of flavonoids in Spatholobus suberectusDunn by impregnation method. Four factor and three level orthogonal design was selectedfrom the first four factors to find best extraction technology of flavonoids of Spatholobussuberctus Dunn based on single factor experiment of ethanol concentration, extractiontemperature, extraction time, solid-liquid ratio and extraction times. According to theresult of orthogonal experiment, the optimal extraction conditions were50%ethanolconcentration,80℃extration temperature,3h extration time and1:30solid-to-liqued ratio.The yield of the flavonoids was7.75%under these conditions.4. Coarse separation of flavonoids in Spatholobus suberectus Dunn. Four kinds ofsolvents such as petroleum ether, chloroform, ethyl acetate and ethyl ether andSpatholobus suberectus Dunn concentrate were used for two-phase solvent extractioningto separate the flavonoids compounds in Spatholobus suberectus Dunn. The experimentalresults showed that: The content of flavonoids compounds in ethyl acetate extractant werehighest, secondly was the ethyl ether extractant, while the content in petroleum ether waslowest. Meanwhile, the results also showed that: The total extraction content of flavonoidin that four extractants was far less than that in50%ethanol extractant, which suggestedthat the main body of flavonoids were still existed in ethanol aqueous solution. 5. Using clearence rate as index, using VCas reference, scavenging avtivities onhydroxyl free radicals and superoxygen anion free radical of six extracts was studied inthis paper. The results showed that clearence rate in different solvent extracts of suberectspatholobus stem for hydroxyl free radicals was50%ethanol extract﹥ethyl acetateextract﹥acetone extract﹥ether extract﹥acetone extract﹥petroleum ether extract; Theclearence rate in different solvent extracts of suberect spatholobus stem for super oxygenanion free radical was50%ethanol extract﹥ethyl acetate extract﹥ether extract﹥chloroform acetate extract﹥acetone extract﹥petroleum ether extract. Comparison on twokinds of free radical scavenging effect showed that the ability of scavenging free radicalsof petroleum ether extract and acetone extract was hydroxyl free radicals﹥superoxideanion free radicals; the order of other extracts to scavenge free radical was consistent, theability of scavenging free radicals was hydroxyl free radicals﹥superoxide anion freeradicals.Moreover, ethanol extract compared with natural antioxidant VCby EC50, thescavenging effect can achieve2times and1.88times to hydroxyl radical and superoxideanion free radical...
