Studies On Extraction, Separation And Bioactivity Of Flavonoids Of Rose

Rose is bud of rose which is rosaceae perennial shrub and it that is a medicinal and edible flower has good food value and health care function. Rose contains a variety of biologically active substances including flavonoids. Preparation conditions of flavonoids of Rose (FOR) were optimized, biological activities were studied and components were analyzed in the paper.1 Water bath extraction and microwave assisted extraction were used to extract flavonoids and each extraction was optimized. The results of water bath extraction showed that the optimal temperature, ratio of solid to liquid, alcohol concentration, microwave extraction time and microwave power were 65℃, 1:45, 70% and 4.5h, respectively, and the extraction ratio for flavonoids is 82.54%. And the results of microwave assisted extraction showed that the optimal temperature, ratio of solid to liquid, alcohol concentration, microwave extraction time and microwave power were 65℃, 1:55, 65%, 45min and 700W, respectively, and the extraction ratio for flavonoids is 82.29%. By comparison the microwave assisted extraction method was selected as the better extraction for FOR in this study.2 Through the static and dynamic adsorption and desorption experiments, FOR purification conditions were optimized in this study. The results showed that the macroporous resins type, sample concentration, flow rate, eluent flow rate, eluent concentration and eluent volume were AB-8 macroporous resins, 3.62mg/mL, 2mL/min, 1mL/min, 70﹪and 2BV, respectively. In this condition the purity of FOR was 32.45﹪and the purity was 6.20 times than before. The yield of flavonoids was 88.67﹪.3 The antioxidant study showed that extraction of acetic ether(EOAE), extraction of n-butanol(EON) , extraction of water(EOW) and purification of AB-8(POA) of the flavonoids had activity of scavenging DPPH·, whose IC50 value were 6.52μg/mL,9.04μg/mL, 6.87μg/mL and 6.34μg/mL, respectively; the inhibitory rate to O2-· of EOAE, EON, EOW and POA whose consentration are 100μg/mL were 14.43%, 27.83%, 23.21% and 21.94%, respectively; the inhibitory rate to·OH of extraction of acetic ether, n-butanol, water and purification of AB-8 whose consentration are 500μg/mL were 23.74%, 43.51%, 32.75% and 27.60%, respectively.4 Three groups of diabetic model mice induced by alloxan were fed respectively with 3 doses of FOR separated and purified by AB-8 macroporous resin for 28d. Then levels of serum glucose(SG) and liver MDA, and liver SOD and GSH-Px activities were investigated. The results showed that by FOR treatment SG and MDA were significantly reduced, and SOD and GSH-Px activities were apparently enhanced. Among 3 doses of FOR treatment, serum glucose levels decrease 26.00﹪in 300mg/ Kg*d which was optimal one. Conclusion: FOR possess significant hypoglycemic activity and can increase the antioxidase activities from liver.5 By Sephadex LH-20 of the initial separation, HPLC separation and UV absorption spectra analysis, at least five flavonoid monomers were contained in Rose and luteolin which was the one of the five flavonoid monomers was identified.
