Object:The flower of Chimonanthus praecoxit is the traditionals plants and the folk medicine, it has many pharmacological functions, using to treat colds, antipyretic analgesic,cough phlegm and lower blood press.The oil from it have stronger antilipidperoxide ability and antibacterial activity,and other biological activities. But the rate of production of essential oil is only0.15%of the total weight. After the extraction of the essential in industrial production,there are plenty of residus. So resulting in a waste of resources.It was reported flavonoid in it have antioxidant activity.If making flavonoids as pharmaceutical raw materials, at the same time in extraction of the essential oil,And increasing its development and utilization, will make the resources fullyutilized.In this article, study on extraction, separation and purificati-on technology of the total flavonoids from the flower of Chimonanthus praecox, monomer components isolating and identifing of monomer components. Also investigating the antioxidant activity of flavonoids. Providing the basis for the further development and utilization of this unique resource.Methods:(1) The smashed flower extracted by steam distillation, saving the essential oil, then water extraction, filtration, the filtrate was concentrated at70℃to medicinal quality and volume of concentrate2:1(g:mL), plus95%ethanol attained to70%ethanol, place48h quietly. The supernatant was concentrated under reduced pressure and concentrate to medicinal quality and volume of1:1(g:mL).(2) Using rutin as reference standard, using UV spectrophotometry to determinthe the total flavonoids from the flower of Chimonanthus praecox,. And conduct methodological test.(3)throught the stable absorption capacity and the eluant effect to select one kind of macroporous resin.(4) By drawing static adsorption and dynamic desorption curve to determine the optimum conditions purification process.(5)Using chromatography on silica gel to separation flavonoid monomers,then using mass spectrometry, nuclear magnetic resonance, infrared spectrophotometry for the structural identification.(6) use Feton theory,conducting experiments oxidation in vitro.Results:(1)The fresh-frozen flowers,grinding,adding purified3times water extracting4h, saving the essential oil, water extraction filtered and the filtrate was concentrated at70℃concentrate to medicinal quality and volume of concentrate2:1(g:mL), plus95%ethanol containing to alcohol of70%,place48h quietly. The supernatant was removed, concentrated at45℃under reduced pressure, recover ethanol, adding water to medicinal quality and the volume ofconcentrate1:1(g:mL).(2) in the study on Determination of Flavonoids in flowers, standard curve were as follows:A=0.0272C-0.0016(A:absorbance, C:concentration: μg/mL) R2=0.9998, at concentration range from0.9mg/L to19.90mg/L has a good relationship.instrument precision RSD=1.0%,stabilize in 60min (RSD=0.88%),so to illustrate the analysis,we need complete experment within Ih, measurement repeatability RSD=0.1%(n=6), the average recovery was100.43%(RSD=0.743%).(3) in the purification process of total flavonoids, select the AB-8macroporous resin, optimum conditions for the purification process were as follows:Taking diluted extract (relative density was0.1),0.5BV amount of resin liquid, taking the sample to AB-8macroporous resin, the optimum rate was2.0mL/min, then the total flavonoids eluted by5BV70%ethanol, elution rate was2.5mL/min, collecting70%ethanol eluent.(4)in the separating of flavonoid monomers dissociation.obtain five pure compounds, the structures of two flavonoids were elucided as:quercetin (Ⅰ) and kaempferol (Ⅱ), another three pending identification.(5) in the study of antioxidant experiment, clear50%of the hydroxyl radical (·OH) solution concentration is required:51.52μg/mL, so it has antioxidant activity.Conclusion:By the study on the extraction, separation, purification of the total flavonoid, obtained the optimum conditions. And the first time to isolate flavonoid monomers, Five compounds were identified,of which two flavone compounds have been elucided. Through the flowers antioxidant experiment shows it has antioxidant activity.
