After considerable development of medical knowledge, a comprehensive knowledge systemshas been formed, people’s needs for medical knowledge and its application have been graduallyimproved, the original means of non-information technology fail to meet the multiple needs ofdifferent groups of people, so the medical information science become an important branch of themedical field. To make effective use of medical resources, realize the sharing and reusing of themedical resources, firstly, we must establish effective mechanisms of knowledge acquisition andmanagement, and build a maintainable, updatable, consistent Domain Knowledge Base, Secondly,achieve the practical application based on the knowledge base.In order to solve the problem of domain knowledge base involved in the Expert AdvisorySystem based on the domain ontology of nephropathy field, this thesis attempts to do a researchfrom the perspective of ontology construction, in view of the the deficiency existing in manual andautomatic ontology construction and the particularity and complexity in the field of medicine.Thenthe thesis proposes a semi-automatic, dynamic update method based on the basis of primitiveontology, and realize the application based on the constructed ontology knowledge base. At first,with the help of the specialist in this field,the thesis defines the key concepts, attributes, relations inthe knowledge system and uses protégéto edit and construct the primitive ontology-Kidonto, anddoes the reasoning experiments,on top of that, first gets the domain knowledge from the dynamicweb text, and then uses the Formal Concept Analysis to construct a new ontology, this dynamicupdate mechanism has been proved effective in the experiment. Finally, the ontology knowledgebase is applied to the actual system,involving the subsystem of question answering which supportsnatural language querying, and can quickly and accurately answer FAQs in nephropathy field, aswell as the subsystem of auxiliary diagnostic which provides preliminary auxiliary diagnosticfunctions, this system also verifies the value of construction of Knowledge Base and the feasibilityof the method in this thesis.
