Herba Dendrobil is sweetness in flavour,slightly cold in nature and acts on the stomach and kidney channels. It have the action of nourishing Yin to clear heat and tonifying the stomach to promoting the production of the body fluid. Modern pharmacology researches show that it can anti-tumor,anti-aging,enhance immunity,etc. Dendrobiutn aurantiacum Rchb.f.var.denneanum (Kerr) Z.H.Tsi is the mainstream breed in Sichuan which quantity is large and curative effect is precise.This paper select it as the object of study. Comparative research between different species and different organs by TLC, HPLC-DAD-ELSD and GC-MS.To provide a scientific basement to prevail the sustainable utilization of Herba Dendrobil and increase breed of it.Part one:Comparative research between D.aurantiacum Rchb. f.var.denneanum (Kerr) Z.H.Tsi and species in Chinese Pharmacopoeia.Objective:Comparative research between D.aurantiacum Rchb. f.var.denneanum (Kerr) Z.H.Tsi, D.nobile Lind1., D.chrysotoxum Lindl. and D.fimbriatum Hook.Methods:1.TLC identification:①Comparative reseach by the methods reference to Chinese Pharmacopoeia;②Systematic reseach on different polarity parts (petroleum ether, ethyl acetate and n-butyl alcohol).2.Determination of active ingredients:③Establish a method for the determination of gigantol by HPLC;②Content determination of tannin compared with gallic acid for the first time;③Assay of the reducing sugar and the total water-soluble polysaccharide content in Herba Dendrobil by spectrophotometric method.Compared with glucose, the water-soluble polysaccharide content was determined.3. Set up the fingerprint chromatograms of chemical principles of Dendrobii Caulis by HPLC-DAD-ELSD for the first time.4. The lipophilic constituents from different spacies were identified by GC-MS.Results:Although they all between to Dendrobium,but there are obvious difference in the inner chemical compositon whether in kind or content. The exist of the same chemical composition, reflects the biological relationships between them. Conclusion:The studies results establish the quality evaluation method. Lay a solid foundation of prevail the application of Herba Dendrobil and increase breed of it.Part two:Comparative study on different organs of D.aurantiacum Rchb.f.var. denneanum(Kerr)Z.H.Tsi.Objective:Comparative study on different organs of D.awantiacum Rchb.f.var. denneanum(Kerr)Z.H.Tsi.Methods:l.TLC identification:systematic study on different polarity parts (petroleum ether, ethyl acetate, n-butyl alcohol).2.Set up the fingerprint chromatograms of chemical principles of different organs of D.awantiacum Rchb.f. var.denneanum(Kerr)Z.H.Tsi. by HPLC-DAD-ELSD for the first time.4. And the chemical composition in the leaf and flower are even more than the stem.Conclusion:Comprehensive consideration, leaf and flower have certain research potential application in addition to stem.
