Objective:To study the chemical composition of bibenzyl compounds from Dendrobinm denneanum; To establish the content determination method for bibenzyl compounds and gigantol; To study the pharmacodynamics effects of immune function, anti-inflammatory, analgesic of bibenzyl compounds from Dendrobinm denneanum.Methods:? Phosphomolybdic acid colorimetric method, Prussian blue and Folin-Denis are commonly used in the determination of bibenzyl compounds, we chose it as the best method which has a suitable maximum absorption. The conditions of the colorimetric method were investigated, then the best condition is based on orthogonal test L9 (34); Using HPLC to detect the content of gigantol.? To study the chemical constituents of bibenzyl compounds from Dendrobinm denneanum. They were purified by column chromatography over silica gel and preparative HPLC.? The experimental mice were randomly divided into five groups. The effects of bibenzyl compounds on immune function of mice were observed by detecting the mice reticuloendothelial cells carbon clearance phagocytice index (K), pinna swelling rate, serum half hemolysis value and weight of immune organ. The anti-inflammatory effects were conducted by xylene-induced ear edema in mice, capillary permeability in abdominal cavity of mice.Results:? Folin-Denis is the method, and colorimetric conditions were as follows:detection wavelength 760 nm, the amount of Folin-Denis 1 ml, the amount of 10% sodium carbonate solution 5 ml, temperature 30 ?,reaction time 1h. Drawing standard curve for bibenzyl compounds and gigantol, respectively:F=0.0780X 0.0878 (r=0.9997), Y=991.72X+10.045 (r=0.9998).? Five compounds were isolated from Dendrobinm denneanum and elucidated as gigantol, syringic acid, vanillin and 5-hydromethylfuraldehyde.? The 72?36mg/kg dose of bibenzyl compounds had obvious growth on mice reticuloendothelial cells carbon clearance phagocytic index. The 72?36?18mg/kg dose of bibenzyl compounds can increase the weight of spleen. And the 72?36? 18mg/kg dose of bibenzyl compounds can increase mice serum hemolysis operation antibody generation. The 72?36?18mg/kg dose of bibenzyl compounds could inhibit the inflammatory response induced by xylene. The 72?36?18mg/kg dose of bibenzyl compounds could reduce the capillary permeability. The 72?36mg/kg dose of bibenzyl compounds could reduce the second phase response of mice methanal test, and the 72?36?18mg/kg dose of bibenzyl compounds can increase mouse threshold.Conclusion:? The method is accurate and simple with good reproducibility, and can control the quality of this preparation effectively.? Bibenzyl compounds were isolated from the part purified by HPD-300 macroporous resin.? Bibenzyl compounds have the effects of immune-regulatory function, anti-inflammatory and analgesic.
