The second largest genus of the orchid is Dendrobium.aurantiacum Rchb.fvar.denneanum (Kerr) Z.H.Tsi is a varietas of D. Aurantiacum Rchb. Herba Dendrobii is homegrown in Sichuan Province. Dendrobium nobile Lindl. and Dendrobium aurantiacum Rchb.f.var.denneanum (Kerr) Z.H.Tsi are the market mainstream varieties in Sichuan and D. aurantiacum Rchb.f.var.denneanum (Kerr) Z.H.Tsi is the most widely distributed Herba Dendrobil in Sichuan. The first record of Dendrobium aurantiacum Rchb.f.var.denneanum (Kerr) Z.H.Tsi is in Shennong’s Classic of Materia Medica, and was classified as high grade in it. It have the action of nourishing Yin to clear heat, tonifying the stomach to promoting the production of the body fluid and moistening lung for arresting cough. Modern pharmacology researches show that it can anti-tumor, anti-aging,enhance immunity, lowering blood glucose, etc. Dendrobium contains several chemical constituents such as alkaloids, dibenzyl, phenanthrene, amino acids, sesquiterpene and polysaccharide.Objective:This research is "Twelfth Five-Year Plan11project of Ministry of science and technology. Taking Dendrobium aurantiacum Rchb.f.vax.denneanum (Kerr) Z.H.Tsi as the study subject. In order to ensure the accuracy of clinical use as drugs, expand the range of applications and increase industry value of Herba Dendrobil, comparative research between D.aurantiacum Rchb. F.var.denneanum (Kerr) Z.H.Tsi and other four species(D.pendulum Roxb., D.ellipsophyllum T. Tang et F. T. Wang., D. Cariniferum Rchb.f, D.trigonopus Rchb. f.) by TLC, HPLC-DAD-ELSD and GC-MS were carried out. We isolate and purify the chemical composition of Dendrobium aurantiacum Rchb.f.var.denneanum (Kerr) Z.H.Tsi to find a suitable index components to set up a determination method.Methods:1.TLC identification:Systematic reseach on different polarity parts (petroleum ether, ethyl acetate and n-butyl alcohol) of five kinds of Dendrobium. 2. Set up the fingerprint chromatograms of chemical principles of five kinds of Dendrobium by HPLC-DAD-ELSD.3. Assay of the reducing Polysaccharides content in Herba Dendrobil by UV-VIS spectrophotometry method for the first time. The spectrophotometry was used with D-glucosum Anhydricum as control.4.The lipophilic constituents from five kinds of Dendrobium were identified by GC-MS.5.This thesis examines the chemical components in the ethyl acetate extract of D.aurantiacum Rchb. F.var.denneanum (Kerr) Z.H.Tsi by the methods of normal phase silica gel, MCI, Sephadex LH-20, preparative thin layer chromatography, recrystallization, ect. HPLC is first utilized in this research and helps to set up an Moscatin content determination method for D.aurantiacum Rchb. F.var.denneanum(Kerr) Z.H.Tsi.Results:1. Although they all between to Dendrobium, but there are obvious difference in the inner chemical compositon whether in kind or content.2. With spectrum and chemical method,7of these compounds could be obtained through separation and purification. All of them have been identified and presented as follows:(1) cis-melilotoside,(2)trans-melilotoside,(3) Naringenin,(4) succinic acid,(5) Moscatin,(6) Melilotoside methyl ester,(7) moscatilin.(6) Melilotoside methyl ester is first separated from D.aurantiacum Rchb.F.var.denneanum(Kerr) Z.H.Tsi.3. HPLC is first utilized in this research and helps to set up an Moscatin content determination method for D.aurantiacum Rchb. f.vai.denneanum (Kerr) Z.H.Tsi, and determine its content.Conclusion:Five kinds of Dendrobium are belong to the same genus but different sections. The similarities and differences in chemical compositions between these Herba Dendrobii reflect the biological relationships between them. In order to control the quality and chemical components of D.aurantiacum Rchb. F.var.denneanum (Kerr) Z.H.Tsi and its compound preparation better, we examines the chemical components in the ethyl acetate extract of D.aurantiacum Rchb. F.var.denneanum (Kerr) Z.H.Tsi.
