Dendrobium aurantiacum Rchb.var.Denneanum(Kerr.)Z.H.Tsi.,also called dieqiaoshihu in Chinese,is the fresh and dry stem of Dendrobium denneanum in Orchidaceae.It has the effect of tonifying stomach and promoting fluid,nourishing Yin and clearing heat.It can be used for deficiency of yin and fluid,thirsty,inappetence,retching,anthemic fever after illness,blurring vision,etc.According to the existing research,Dendrobium denneanum contains many kinds of compounds such as polysaccharides and bibenzyls.The former has the effects of improving immunity and hypoglycemic action,the latter has the effects of antitumor,antioxidation and other pHarmacological effects.Dendrobium denneanum is listed in the Standards of Traditional Chinese Medicine in Sichuan Province(2010 edition)and the Processing Standards of Traditional Chinese Medicine in Sichuan Province(2015 edition).It is a genuine medicinal material in Sichuan Province.It is a genuine medicinal material in Sichuan Province and has been planted on a large scale.At present,most of the pHarmacopoeia and local standards do not specify the harvest time of Dendrobium.Because wild Dendrobium resources were scarce in the past,the description of harvestable throughout the year was applicable to the situation of wild scattered plants,also called "Harvest as you see".Nowadays,many kinds of Dendrobium have been planted on a large scale.In order to ensure the quality of medicinal materials and Large-scale cultivation of Dendrobium,it is urgent to carry out the related research on the harvest period of Dendrobium.it is urgent to makes relevant research on the harvest period of Dendrobium.In addition,there are many flexible fibers in Dendrobium,and the traditional primary processing method is cutting into segments.With the development of cutting technology,cutting into slice is feasible,the cutting method has a new choice.The advantages of using slice need to be validated.Based on the above purposes,this paper carried out the research on the suitable harvesting time of Dendrobium denneanum and the comparative study of slice cutting.So we makes the research of the best harvest time and the comparation of slice and segment of Dendrobium denneanum.The specific contents are as follows:1.In order to measure the quality of Dendrobium denneanum,the corresponding indicators were established in this paper.(1)The determination method of polysaccharide content was established and the uncertainty was evaluated.Referring to the content of determination method of Dendrobium officinale polysaccharide content in Chinese pHarmacopoeia(2015 edition),a method for the determination of polysaccharide content in based on phenol-sulfuric acid method was established.The methodological investigation showed that the method had good repeatability,strong stability and high accuracy,and was suitable for the determination of polysaccharide content in Dendrobium denneanum.In order to improve the accuracy of the method,GUM method and MCM method were used to analyze the uncertainty of the established method.The 95% probability of the determination results of polysaccharide content in Dendrobium denneanum was calculated by MCM method,and the inclusion interval was [15.28%,16.16%].The results showed that the method was accurate and reliable.The GUM method has not been validated,its description of uncertainty components is still referable.(2)The existing methods for the determination of total bibenzyls were optimized,and a method for the determination of Dendrobium phenol was established.On the basis of the total amount of bibenzyls in our research group,the method was optimized,some shortcomings of the original method were improved,the sensitivity and stability of the method were improved,and the methodological investigation showed that the method was stable and accurate.A method for the determination of bibenzylpHenols in Dendrobium benzylides was established,and the methodological investigation showed that the method was stable and accurate.(3)Cis-melilotoside was isolated and identified from Dendrobium denneanum.A method for the determination of cis-melilotoside was established.The content of cis-melilotoside was separated from Dendrobium denneanum by macroporous resin column chromatograpHy and the reference substance has made.The corresponding content determination method was established.Through methodological investigation,its showed that the method was stable and accurate.(4)The fingerprint method of Dendrobium denneanum by HPLC was established.A method for fingerprint analysis of Dendrobium denneanum by HPLC was established,and methodological investigation was showed that the method was stable and accurate.Eleven batches of Dendrobium denneanum samples were determined.The result was analyzed by principal component analysis,cluster analysis and thermal map.(5)The quality of Dendrobium denneanum was comprehensively measured by other indexes such as drying rate,water absorption rate and extract.The yield of medicinal materials was measured by drying rate,the permeability of different forms of medicinal materials in water was measured by water absorption rate,and the dissolution efficiency of different forms of medicinal materials was measured by extracts.2.The suitable harvest time of Dendrobium denneanum was determined by comprehensive analysis of each index and TOPSIS method.The contents of 11 batches of Dendrobium denneanum samples collected at different harvesting times were determined by the established content determination method.The results showed that the drying rate was higher in February of each year and the highest in spring of the second year.Dendrobium phenol content increased with the growth period.The polysaccharide content was higher in spring and lower in autumn in the second year.Based on the variation of bibenzyls and glycosides,fingerprint information was analyzed by various statistical methods.The growth of Dendrobium sheathii could be divided into three stages by taking November of the first year and July of the third year as nodes.In the second winter to the early spring of next year,DQ8 batches of Dendrobium denneanum stem(DQ8 batches)had higher content of components,which would be suitable for harvesting.The results of the comprehensive evaluation of fingerprint by using entropy weight method and TOPSIS method are the same.3.The Decoction model of simulated clinical application and the warm soaking model of simulated daily tea drink were established.The advantages and disadvantages of the slices and segments of Dendrobium denneanum were compared by using the established index determination methods.After steaming,Dendrobium denneanum was cut into 10 mm segments and 4 mm slices.The conditions of decoction for 60,90,120 minutes and warm soaking for 60,90 minutes were simulated respectively.The polysaccharide dissolution,phenol dissolution,total bibenzyls,water absorption and extract were determined.The water absorption rate of the slice of Dendrobium denneanum was 1.46 times higher than the segment;about the water-soluble extract,the slice were 1.2 times higher than the segment;the alcohol-soluble extract,the slice were 1.1 times more than that of the segment;the average polysaccharide extraction rate of the slice was 1.17 times higher than the segment,the average bibenzyl component extraction rate of the slice was 1.3 times higher than the segment,and the pHenol extraction rate of the slice was 2.5 times higher than that of the slice in the simulated warm-leaching process.Compared with segment polysaccharides,the average dissolution rate of polysaccharides and bibenzyls with cutting into slice increased by 1.11 times and 1.5 times respectively.Slices could significantly improve the dissolution rate of components.
