Objective To determinate the effect of Shenfuqiangxin(SFQX) decoction in chronic heart failure(CHF) rats model. To observe the influence of SFQX decoction on plasma AVP and urinary AQP-2in CHF rats, in order to explore mechanism of SFQX decoction on water retention in CHF rats model.Methods Using male adult SD rats made CHF models by coarctation of abdominal aorta (CAA), gave the echocardiogram4weeks later to appraise whether the cardiac function of operated rats got worse compared with sham-operated rats.60successfully established CHF rats were randomly divided into5groups:large-dose group, medium-dose group, low-dose group, Losarton group, and model group. Large-dose group (14g/kg·d, SFQX decoction pretreatment), medium-dose group (7g/kg·d, SFQX decoction pretreatment), low-dose group (3.5g/kg·d, SFQX decoction pretreatment), Losarton group (10mg/kg·d), sham group and model group(2ml/d sterile purified water pretreatment). At the end of4weeks, cardiac function was measured by ultrasonic echocardiogram, plasma AVP and urinary AQP-2were detected by ELISA method.Results Comparing with model group, the common condition and cardiac function were improved. The concentration of plasma AVP and urinary AQP-2in each dose SFQX decoction were reduced(p<0.01). The concentration of plasma AVP and urinary AQP-2reduced correspondingly with the increase of the dose of SFQX decoction, while ejection fraction(EF) reducing. There was a remarkable negative correlation between EF and the concentration of plasma AVP and urinary AQP-2.Conclusion SFQX decoction can improve the cardiac function and reduce the concentration of plasma AVP and urinary AQP-2in CHF rats model, which is in a dose-dependent manner. The mechanism of SFQX decoction on water retention in CHF rats may be related to reduced concentration of plasma AVP and urinary AQP-2.
