BackgroundCervical cancer is a malignant tumor occurred in the cervical epithelium. Incidence of cervical cancer is second only to breast cancer in women, accounting for more than half of the female reproductive system tumor, has been a serious threat to women’s physical and mental health. In recent years, there is a rising trend in mortality rate, new cases each year about529000people around the world, about140000new cases in China. However, the occurrence of cervical cancer is a complex process of more genetic level change, and related to a variety of oncogenes and tumor suppressor genes expression, regulation and apoptosis. Currently, molecular biology research is to clarify the reasons for this loss of control. Oncogene is mostly required for normal cell growth and growth factors receptors, as a result of mutation, amplification and rearrangements that excessive cell growth; additional lack of some tumor suppressor genes, loss, damage or variant inactivation balanced regulation of cell growth, the cells lose their normal control patients with tumors or cause to promote tumor progression.Steroid receptor activation factor3auxiliary (steroid receptor coactivator-3, SRC-3) SRC-3belongs to the nuclear hormone receptor activator total P160members of the family, the expression of the protein is a kind of the nuclear hormone receptor transcription activating factor, is one of the newly discovered in recent years proto-oncogene. F box/wd-40domain protein7(F- box/WD repeat-containing protein7, FBXW7) belongs to the F-box members of the family of proteins, is SCF ubiquitin ligase target protein identification component. Recent studies have shown that FBXW7is a tumor suppressor, the role of many substrates is oncogene, FBXW7deletions and mutations can lead to human rapid tumor growth. At home and abroad for the SRC-3, FBXW7expression in cervical cancer research is relatively rare.ObjectiveResearch steroid receptor coactivator factor3(SRC-3), F box/WD-40domain protein7(FBXW7) two proteins in cervical cancer development process and to analyze them with various clinical and pathological features of cervical cancer mutual relations and further diagnosis and treatment of cervical cancer provide new targets and new ideas.Materials and Methods1Source of specimen Select our hospital first affiliated hospital of zhengzhou university in June2011-June2012gynecological surgery patients, cervical cancer group, a total of60cases, clinical data complete, by the department of pathology, the first hospital of paraffin embedding, making slice. All cervical cancer patients aged between24and68, the average age was (44.80±10.92), has not been received preoperative radiotherapy or chemotherapy, postoperative pathological diagnosis. By organization type points:48cases squamous cell carcinoma, adenocarcinoma,12cases by FIGO stage (2000):Ⅰ-Ⅱ-a period of32cases, Ⅱ b-Ⅳ28cases; According to the degree of tumor differentiation points:high, medium and low differentiation of16cases,20cases,24cases respectively; According to the presence of lymph node metastasis:no lymph node metastasis was34cases, and26cases with lymph node metastasis. And at the same time for other reasons the cervical resection of20cases of normal cervical tissues as control.2Immunohistochemical SP method detected SRC-3, FBXW7in cervical cancer and normal cervical tissues, analysis of these two indicators and cervical cancer clinicopathological features as well as the relationship between the correlation that exists between them, and then discuss the role of both in the process of development of cervical cancer.3Statistical methods Using SPSS17.0statistical software for statistical analysis, using X2test method to analyze the same indicator with different clinical and pathological features of cervical relationship between both methods using Spearman rank correlation analysis the relationship between SRC-3, FBXW7two indicators expressed in cervical cancer, the test level (α)=0.05.Results1The positive expression rate of SRC-3in cervical cancer tissue was33.3%, and the positive rate in normal cervical tissues was10%.there was no statistically significant difference between them. The expression of SRC-3protein in cervical cancer was closely related with tissue differentiation, clinical stage, lymph node metastasis, but not with age, histological type was no significant correlation.2The positive expression rate of FBXW7in cervical cancer tissue was48.3%, and the positive rate in normal cervical tissue was90%, the difference was statistically significant. FBXW7protein expression was closely related cancer tissue differentiation, clinical stage, lymph node metastasis, but not with age, histological type was no significant correlation.3There is a negative correlation by the Spearman rank correlation analysis between the expression of FBXW7and SRC-3two indicators in cervical cancerConclusions1The expression of SRC-3protein in cervical cancer were significantly higher than in normal cervical tissue and related with cancerous tissue with depth of invasion, lymph node metastasis and clinical stage, indicating that SRC-3overexpression in the development of cervical cancer process promotes cancer invasion and metastasis.2The expression of FBXW7in normal cervical tissues was much higher than cervical cancer tissues, and closely related with differentiation degree, lymph node metastasis and clinical stage of cervical cancer, indicating that low expression of FBXW7protein in cervical cancer development process enhanced malignancy and metastasis.3There is a remarkable correlation between SRC-3and FBXW7two indicators in cervical carcinoma. The abnormal expression of SRC-3and FBXW7might play a vital part in the process of development of cervical cancer. Combined detection of SRC-3, FBXW7two indicators will help predict of cervical cancer progression and metastasis, they also may be in the early diagnosis of cervical cancer, cancer prognosis, and provide important reference value to find new targets for molecular targeted therapy.
