Objective: To research the expression of EZH2、BMI-1、FBXW7 in esophageal squamous cell carcinoma and adjacent normal tissue, and explore EZH2、BMI-1、FBXW7 relevance to neoadjuvant chemoradiotherapy in the near future curative effect.Methods: Select a large number of esophageal squamous cell carcinoma and adjacent normal tissues which were diagnosed by endoscopic biopsy in the first affiliated hospital of Henan University of Science and Technology from 2013.01 to 2014.05. 30 cases of cancer and adjacent normal tissues were selected from which were first diagnosed and didn’t undergo neoadjuvant chemoradiotherapy, and the expression of EZH2, BMI-1 and FBXW7 were analyzed by immunohistochemical technique in these patients. All patients were detected by barium meal and CT before and after treatment. Prospective analysis of the expression levels of EZH2, BMI-1, FBXW7 and the relationship with term efficacy of neoadjuvant chemoradiation therapy.Results: 1. The positive expression of EZH2 in esophageal squamous cell carcinoma 60%(18/30) was higher than in adjacent normal tissue 30%(9/30), with statistical significance P = 0.020(P <0.05); The positive expression of BMI-1 in esophageal squamous cell cancer 63.3 %(19/30) was higher than in adjacent normal tissues 26.7%(8/30), P = 0.004(P <0.05), there was statistically significant; The positive expression of FBXW7 in esophageal squamous cell carcinoma 40%(12/30) was lower than in adjacent normal tissue 80%(24/30), P = 0.002(P <0.05), there was statistically significant; 2. The positive expression of EZH2, BMI-1, FBXW7 in esophageal squamous cell carcinoma of the short-term efficacy of neoadjuvant chemoradiotherapy. EZH2 expression positive patients with efficient 44.4%(8/18) in esophageal squamous cell carcinoma patients but with negative expression efficiency of 66.7%(8/12), P=0.284(P>0.05), there was no statistically significant; BMI-1 expression positive patients with efficient 36.8%(7/19) in esophageal squamous cell carcinoma patients but with negative effective rate 81.8%(9/11), P=0.026(P<0.05)there was statistically significant; FBXW7 positive expression efficiency is 75%(9/12) in esophageal squamous cell carcinoma patients, but the expression of negative effective rate 38.9%(7/18), P=0.072( P > 0.05), there was no statistically significant; 3. In exploring EZH2, BMI-1, FBXW7 with esophageal squamous cell carcinoma of the short-term efficacy of neoadjuvant chemoradiotherapy and Logistic regression analysis, to neoadjuvant chemoradiotherapy as the dependent variable, the age, sex, length of tumor, tumor location, the EZH2, BMI-1, FBXW7 expression of high and low as independent variables in Logistic regression analysis. The results showed that independent factors of esophageal squamous cell carcinoma chemoradiotherapy is EZH2 high expression(P=0.032), BMI-1high expression(P=0.031), FBXW7(P=0.041) low expression, the difference was statistically significant; 4. The positive expression of EZH2 protein with the positive expression of BMI-1 is a significant positive correlation in ESCC(r=0.367, P=0.046); The positive expression of BMI-1 and FBXW7(r=0.198, P=0.295), EZH2 and FBXW7(r =-0.028, P=0.884) were not relevant in ESCC.Conclusion: 1. The positive expression of EZH2 and BMI-1 protein in ESCC were higher than in adjacent normal tissue, and the positive expression of FBXW7 in ESCC was lower than adjacent normal tissues, which suggested that EZH2, BMI-1, FBXW7 expression may be associated with abnormal esophageal cancer occurrence and development.2. The positive expression of BMI-1 protein in ESCC which was lower than negative expression in Neoadjuvant chemoradiotherapy short-term effect. But the positive expression of EZH2 and FBXW7 in ESCC were no statistically significant in in Neoadjuvant chemoradiotherapy short-term effect. It showed that the positive expression of BMI-1 may predict neoadjuvant chemoradiotherapy short-term effect in esophageal squamous cell carcinoma.3. The high and low expression of EZH2、BMI-1 and FBXW7 may be an independent factor in ESCC recent neoadjuvant chemoradiotherapy treatment; Showed that to detect the high and low expression of EZH2、BMI-1 and FBXW7 may be effective to predict neoadjuvant chemotherapy short-term efficacy of the reference index.4. The positive expression of EZH2 and BMI-1 protein was a significant positive correlation. Showed that they might be involved in the development、invasion and metastasis of interaction and participation in esophageal tumor cells.
