Objective: The purpose of this study was to investigate the expression of F-Box and WD40 domain protein7(FBXW7),Hypoxia-inducible factor 1 alpha(HIF-1α)and endase-1(EN01)in laryngeal squamous cell carcinoma(LSCC)and analyze the roles and relationship of them in the genesis and development of laryngeal squamous cell carcinoma(LSCC)according to their clinical pathological datas,and then to explore their clinical significance..Methods: 41 cases of laryngeal squamous cell carcinoma samples treated in the department of otolaryngology of the second hospital of hebei medical university from October 2016 to October 2017 were collected.All pateints received no chemoradiotherapy treatment.All specimens were pathologically confirmed as squamous cell carcinoma.Clinical stages were conducted according to TNM classification standards published in 2002 by the international association against cancer(UICC).Meanwhile,we collected normal adjacent tissues as control group.The expression of FBXW7,HIF1 proteins and ENO1 in laryngeal squamous cell carcinoma(LSCC)and adjacent tissues were detected by streptavidin peroxidase(SP)immunohistochemistry The experimental datas were statistically analyzed using SPSS 22.0.The positive protein expression rate was analyzed by chi-square test,and the correlation between the expressions of FBXW7,HIF-1 and ENO1 in laryngeal cancer tissues was assessed by Spearman level correlation analysis.P<0.05 was taken as statistically significant difference.Results:1.The positive expression rate of FBXW7 was 24.39% and 90.24%,respectively,in laryngeal and paracancerous tissues,with P<0.05,which was statistically significant.The positive expression rates of FBXW were 34.62% and 6.67% in the high differentiation group and the medium-low differentiation group,respectively,with P<0.05,showing statistical significance.The positive expression rates of FBXW7 were 33.33% and 7.14% in those with no lymph node metastasis and with lymph node metastasis,with P<0.05,which was statistically significant.The positive expression rates of FBXW7 in stage I~II and stage III~IV laryngeal cancer were 30.00% and 19.05% respectively,and P>0.05,which was not statistically significant.The positive expression rates of FBXW7 were 31.58% and 18.18%,respectively,in patients aged less than 60 years old and more than 60 years old,respectively,P>0.05,without statistical significance.The positive expression rates of FBXW7 were 31.82% and 15.79%,respectively,in patients with laryngeal cancer whose smoking volume was no more than or equal to 20 sticks/day and more tha 20 sticks/day,respectively,with P>0.05,which was not statistically significant.2.The positive expression rates of HIF-1 were 75.61% and 21.95% in laryngeal and paracancerous tissues,respectively,P<0.05,which was statistically significant.The positive expression rates of HIF-1 were 86.67% and 69.23% in the medium-low differentiated cancer tissue and high differentiated cancer tissue,respectively,P<0.05,showing statistical significance.The positive expression rates of HIF-1 in stage III-IViv and I~ II were 80.95% and 70.00%,respectively,P>0.05,without statistical significance.The positive expression rates were 100.00% and 62.96%,respectively,in the lymph node metastatic group and the lymph node non-metastatic group,P<0.05,which was statistically significant.The positive expression rates of HIF-1 were 68.42% and 81.82%,respectively,at the group of pateints less than or equal to 60 years old and the group of patients more than 60 years,respectively,P>0.05,without statistical significance.The positive expression rates of HIF were 81.82% and 68.42% in patients with daily smoking of no more than 20 cigarettes and more than 20 cigarettes respectively,P>0.05,without statistical significance.3.The positive expression rates of ENO1 in laryngeal cancer tissues and adjacent tissues were 80.49% and 21.95%,respectively,P<0.05,with extremely statistical significance;The positive expression rates of ENO1 were 73.08% and 93.33%,respectively,in the highly differentiated laryngeal cancer group and the medium-low differentiated cancer group,P<0.05,which was statistically significant.The positive expression rates of ENO1 were 74.07% and 92.86% in patients without lymph node metastasis and patients with lymph node metastasis,respectively,P<0.05,which was statistically significant.The positive expression rates of ENO1 in stage I~ II and stage III~IV laryngeal cancer groups were 75.00% and 85.71%,respectively,P>0.05 which was not statistically significant.The positive expression rates of ENO1 were 78.95% and 81.82% in the laryngeal cancer groups aged less than or equal to 60 years old and more than 60 years old,respectively,with P>0.05,which was not statistically significant.The positive expression rates of ENO1 were 81.82% and 78.95%,respectively,in the laryngeal cancer group whose smoking volume was no more than 20 cigarettes per day and more than 20 cigarettes per day,respectively,with P>0.05,showing no statistical significance.4.Spearman rank correlation was used to analyze the counting data of FBXW7,HIF1 spread and ENO1,and it was found that the correlation coefficients were-0.553,-0.396 and 0.580,respectively,which had statistical significance(P<0.05).Conclusion:1.The decreased expression of FBXW7 was related to the occurrence of laryngeal cancer,the pathological differentiation degree and lymph node metastasis of laryngeal cancer,and it was not related to the clinical stage of laryngeal cancer,the age of patients and daily smoking amount.2.The increased expression of HIF-1 and ENO1 were associated with the occurrence of laryngeal cancer.It was not associated with clinical stages,the age of patients,and daily smoking amount,however,it was related to pathological differentiation degree and lymph node metastasis.3.HIF-1 and ENO1 might pass through the negative feedback loop of FBXW7 to increase the glycolytic activity and then reduced the level of tumor suppressor genes.The results of this study suggested that FBXW7,HIF-1 and ENO1 could be used as potential biomarkers to provide reference for the clinical diagnosis,screening and prognosis of laryngeal cancer.Specific expressed inhibitors of FBXW7 and HIF-1 and specific expressed agonist of ENO1 might be a starting point for the research of anti-tumor drugs in individualized treatment,providing new ideas for improving the treatment effect of laryngeal cancer and inhibiting the progression of laryngeal cancer.
