Objective:To explore the effects of genistein on sex hormone,on Ovary and uterus in rats with polycystic ovarysyndrome. To make sure the effects of genistein in rats with polycystic ovary syndrome. To evaluate theexpressing of cytochrome P450aromatase on Ovary and uterus in rats with polycystic ovary syndrome, andits effection mechanisms.Methods:Making animal model at insulin (INS) and HCG, drive the rats into6groups: the control group,the modelgroup, the low dosed Gen group, the middle dosed Gen group,the high dosed Gen group and the estrogengroup.Feeding5mg/(kg·bw·d)GEN for the rats in low dosed Gen group, Feeding10mg/(kg·bw·d)GEN for the rats in middle dosed Gen group, Feeding20mg/(kg·bw·d)GEN for the rats in high dosed Gengroup.Feeding0.5mg/kg diethylstilbestrol for the rats in estrogen group;Feeding the GEN anddiethylstilbestrol for15days. At the end of the testion, observe the rats in Oestrous cycle for10days.Afer thelast drugs feeding,stop feed the rats food but no stop the water. Weighing all the rats in electronic balances.Then kill the rats for the blood with will used for which is used in ELISA method to detect hormone levels;Stripping rat ovary and uterus, side ovarian fixation, HE staining, slicing, used for electron microscopeobservation, the other side of the ovary and uterus liquid nitrogen preservation, used for RT-PCR method todetect P450aromatase expression in determination.Results:(1) Modeling test cuhk mouse vaginal epithelial cells smear two consecutive personality cycle (a total of10days, each period of5days), model group vaginal keratinocytes continue to appear. Model group comparedwith blank control group, body weight fell slightly, but not obvious.(2) Compared with model group, the medium and high dose group and estrogen group rats serum P levelrise, the extremely significant difference (P <0.01). Low dose group of rats serum T level drops, significantdifference (P <0.05); Medium and high dose group and estrogen group T levels drop, significantly significantdifference (P <0.01). Dosage compared with model group, each group of rats serum SHBG levels areincreased, and extremely significant.(3) Compared with model group, each dose group rats ovaries area and volume decreased, the decline inestrogen group was obviously, there is significant difference (p <0.05). Compared with model group, middle,high dose group of rats uterus/body ratio increased significantly, with significant difference (p <0.05), ovarianestrogen group/body ratio decreased significantly, with significant difference (p <0.05). The microscopicobservation of visible, model group rats appear ovarian PCOS symptoms of morphological changes, confirmedtest building success; Dose group rat ovarian morphology changed, especially in high dose group (20mg/kgbw) changed obviously, compared with model control group, high dose group GEN rat ovarian theca cell layeris thinning model control group, granular cell layers increased significantly, the configuration is complete,arrangement is neat, and corpus luteum organization number. (4) Compared with model group, the estrogen group and high dose group of PCOS rats uterus andcytochrome P450aromatase expression in ovarian tissue level is significantly reduced. Compared with modelgroup, middle dose group and low dose group of PCOS rats uterus and ovarian tissue of cytochrome P450aromatase expression level difference is not obvious.Conclusion:(1)Genistein can play an important role on Polycystic Ovary Syndrome.(2)May be P450aromatase has an important role in Polycystic Ovary Syndrome to study thepathogenesis of PCOS disease provides a new direction and clues.
