| Objective:Study the iron metabolism and iron nutrition state in obese rats,observe thechanges of iron related protein’s mRNA expression,and provide the evidence for theprevention and control of iron’s deficiency induced by obesity.Methods:30male wistar rats were divided into control group(n=10)and obesitygroup(n=20).The rats were fed with ordinary feed, high fat diet,respectively for12weeks,then they were killed by taking the eye blood and collected the blood.Thehemoglobin,red cell count,hematocrit,serum iron,total iron binding capacity,serumtransferrin saturation,hepcidin,transferring and ferritin of rats were determined.Determine the mRNA espression of liver indicators,included ferritin,transferrin,hepcidin,small intestine divalent metal transporter, transferrin receptor,adipose tissueleptin.Results:Compared with control group, the red cell count and hematocrit value in obesitygroup was increased,and the hemoglobin content was decreased,compared withcontrol group showed significant differences(P<0.05).The serum iron,iron saturation values in obesity group were decreased,the serumtotal iron binding capacity was increased,compared with control group showedsignificant differences(P<0.05).The ferritin,transferrin and hepcidin values in obesitygroup were reduced,compared with control group showed significant difference (P<0.05).Compared with the control group,liver ferritin’s mRNA expression increasedsignificantly in obese group,and the difference showed statistical significance (P<0.05),hepcidin’s mRNA expression was reduced significantly,and the differenceshowed statistical significance(P<0.05),transferrin’s mRNA expression was increasedsignificantly, and the difference showed statistical significance(P <0.05).Compared with the control group,obese rats’ small intestine divalent metaltransporter was reduced,transferrin receptor’s mRNA expression was increasedsignificantly,and the difference showed statistical significance(P <0.05).Compared with the control group, obese rats’ adipose tissue leptin’s mRNAexpression was increased significantly,and the difference showed statisticalsignificance(P <0.05).Conclusion:1. Obesity can cause the body iron metabolism change, appear iron deficiency.2. Obese rats’ liver ferritin’s mRNA expression, transferrin’s mRNA expressionwas increased; hepcidin’s mRNA expression was reduced.3. Obese rats’ small intestine divalent metal transporter’s mRNA expression wasreduced;transferrin receptor’s mRNA expression was increased.4. Obese rats’ adipose tissue leptin’s mRNA expression was increased. |
