Objective: Study the calcium metabolism state changes in obesity rats,observe the effect of calcium supplementation on obesity,and provide a theoretical basis for the further study of the relationship between obesity and calcium.Methods: In accordance with the principle of ra ndomization,40 male Wistar rats were divided into control group(n=10)and obesity group(n=30).The rats were fed with normal diet and high fat diet,respectively for 8 weeks.Then all ten rats in control group and the randomly selected ten rats in obesity group were killed.Detect the following indicators: serum calcium,alkaline phosphatase,parathyroid hormone, calcitonin,vitamin D receptor.Calcium binding protein,25 hydroxy vitamin D. Secondly,the remaining 20 rats were used in the next part. The remaining 20 male Wistar rats were divided into obese control group(n=10)and calcium group(n=10). All the rats were fed with normal diet. The rats were lavaged with calcium gluconate solution,normal saline,respectively for 15 days,then they were killed. Detect the following indicators: lipids four(CHO,TG,LDL-C,HDL-C),osteocalcin,25 hydroxy vitamin D.Determine the gene expression of leptin and fatty acid synthase by Real-time PCR.Results: 1.Compared with control group,the serum calcium in obesity group was decreased,alkaline phosphatase in obesity group was increased,and both of them showed significant differences(P<0.05). 2.Compared with control group,the parathyroid hormone in obesity group was increased,the calcitonin in obesity group was decreased,and both of them s howed significant differences(P<0.05).The calcium binding protein in obesity group was decreased,the vitamin D receptor was increased,the 25 hydroxy vitamin D was decreased,all of them showed significant differences(P<0.05). 3.Compared with control group,the calcium binding protein gene expression and the vitamin D receptor gene expression bothdecreased in obesity group,and both of them showed significant differences(P<0.05). 4.Compared with obese control group,the weight and Lee’s index in calcium group both were decreased,and both of them showed significant differences(P<0.05).Total cholesterol,triglyceride and low density lipoproteinthe in calcium group were decreased,and all of them showed significant differences(P<0.05).High density lipoprotein in calc ium group were increased,and showed significant differences(P<0.05). 5.Compared with obese control group,the osteocalcin and 25 hydroxy vitamin D in calcium group both were increased,and both of them showed significant differences(P<0.05). 6.Compared with obese control group,the leptin and fatty acid synthase gene expression decreased in calcium group,and both of them showed significant differences(P<0.05).Conclusion: 1.Obesity can lead to the serum calcium decrease and alkaline phosphatase activity increase in rats. 2.Obesity can lead to the rat serum parathyroid hormone level increase, the level of calcitonin reduce, the level of calcium binding protein reduce, the level of vitamin D receptor reduce, the level of 25 hydroxy vitamin D reduce. 3.Obesity can lead to the gene expression of calcium binding protein and vitamin D receptor reduce in small intestine tissue. 4.Calcium can cause body weight, Lee’s index decrease, total cholesterol, triglyceride,low density lipoprotein decrease,high density lipoprotein increase.5.Calcium can cause rat serum osteocalcin and 25- hydroxy vitamin D level increase. 6.Calcium can cause the gene expression of leptin and fatty acid synthase reduce in white adipose tissue.
