Objective:To investigate the influence and mechanism of weanling rats' iron-fortified diet to adulthood obesity induction. Methods:85 male SD weanling rats were divided randomly into basic diet group, high (4 times), medium (3 times) and low (2 times) dose iron-fortified group. After 4 weeks of iron-fortified intervention, all rats were fed with basic diet for 1 week. After intervention, blood glucose, serum lipid, blood iron, the fat/body ratio and the levels of leptin and insulin were compared. We took livers and kidneys for pathological studies by HE dyeing. Subsequently, the remaining basic diet group's adult rats we divided randomly into normal controls and obesity induction group. Obesity induction group and three iron-fortified groups were all fed with high-fat diet. After 8 weeks of obesity induction, all rats were put to death and tested in the indexes which mentioned above, and adipose tissue of high-testosterone weeks and kidney weeks were weighed for calculating the fat/body ratio, at the same time, a reverse transcription polymerase chain reaction (RT-PCR) technique was used to analysis the expression of UCP2, UCP3 and GLUT4 mRNA in muscle. Results:1. After the Iron-fortified intervention, the weight of the medium dose iron-fortified group was lower than other groups; After obesity induction, weight, weight growth and fat/body of obesity induction group were higher than other groups; medium dose iron-fortified group's weight was lower than normal control group; 2. After the Iron-fortified intervention, high iron-fortified group' blood glucose content was lower than basic diet group; After obesity induction, TG and TC content of obesity induction group were significantly higher than other groups; TC content of three iron-fortified groups were slightly higher than normal control group; the HDL-C leves of medium dose iron-fortified group and normal control group were higher than high dose iron-fortified group and obesity induction group; 3. After the iron-fortified intervention, the leptin and insulin level of three iron-fortified groups were higher than basic diet group; After obesity induction, the leptin and insulin level of obesity induction group were significantly higher than other groups, three dose iron-fortified groups'leptin level were higher than normal control group, the insulin level was lower than normal control group; 4. The expression level of UCP2, UCP3 and GLUT4 mRNA of obesity induction group were significantly lower than other groups, medium dose iron-fortified group's expression level was higher than other groups; The expression level of UCP3 and GLUT4 mRNA of high dose iron-fortified group were lower than normal control group. Conclusion:Appropriate iron supplement in rats' early life can continue affecting adulthood obesity formation induced by high-fat feed, increase sensitivity to leptin, prevent insulin resistance, increase the expression level of UCP2, UCP3 and GLUT4 gene.
