Objective:To investigate the expression of DC-SIGN on the dendritic cells in spontaneousabortion and the possible role in the mechanism of spontaneous abortion.Methods:During2011.6-2011.9, we collected decidual tissue of gestational age8-10weekspregnant in the Department of gynecology and obstetrics factors for spontaneous abortionor voluntary family planning factors curettage or abortion. Natural abortion group criteria:No genetic disease, endocrine, genital anatomical abnormalities, tumor, immune diseasehistory; no long-term medications; patients without history of spontaneous abortion, or thefirst pregnancy abortion; ultrasonography prompted to the embryo stops growing. Thecontrol group included: color Doppler prompt visible fetal heart fetal bud, no previoushistory of abortion, or for the first pregnancy. Immunofluorescence and RT-PCR to detectDC-SIGN expression in the decidual tissue. compared with group of spontaneous abortionand family planning group expression and distribution differences. statisticallytested using SPSS13.0Software, P <0.05for the difference was statistically significant.Results:1By immunofluorescent technique for detection of visible natural abortion group andcontrol group with DC-SIGN expression, mainly distributed in the membrane of the cell.using ImageJ software to analyze the number of positive cells, respectively:46.20±2.40and78.21±0.28, spontaneous abortion group in the DC-SIGN+DCs express less than thecontrol group, and there was significantly different (t=63.03, P <0.01).2By using the techniques of RT-PCR test results show that: the group of spontaneousabortion decidua of normal DC-SIGN mRNA relative expression was significantly lessthan that in the control group, and there was statistical significance (P=0.02, P <0.05).Conclusions:Dendritic cells (DC) play an important role in the maternal-fetal immune tolerance mechanisms, and expression of DC-SIGN decrease in the occurrence of spontaneousabortion may play a certain role. This experiment may provide new ideas for spontaneousabortion mechanism to provide a new theoretical ideas.
