Objective:Through the study the impact of the xiaoyan powder on the analgesic effect in mice and the active of intercellular adhesion molecule1in the gouty arthritis synovia of rat,to discuss some of the mechanism and effect of the xiaoyan powder in orthopaedic.Methods:The analgesic experiment method:the hot-plate experiment:choose30healthy BALB/C female mice with pain threshold from5seconds to30seconds,divide them into3groups in accordance with the random permutation table, each group has10ones.they are Xiaoyan powder group,Futalin group, normal group. The Xiaoyan powder group uses the xiaoyan powder,the Futalin group applies futanlin cream on the mice’s feet,then fix the claws with the infusion paste. The normal group uses the distilled water.At last,test each group’s pain threshold after1hour, the writhing experiment: choose ICR30healthy mice (male and female’s number are equal).Specific groups and medication are the same as the hot-plate experiment.After60minutes, inject0.6%acetate0.1ml/10g into intraperitoneal;observe the mice’s writhing frequency in15minutes after administration.Gout arthritis drug intervention experiment:choose40healthy male SD rats with weight from200g to220g; divide them into4groups in accordance with the random permutation table, each group has10rats. Group A is Xiaoyan powder group; group B is Futalin group (western medicine group); group C is model group; group D is normal group. Make the gout arthritis model in the right ankle of group A, Band C. Use6th needle from the joints of the leg back to the right side from the direction of45degrees centigrade into medial tibial tendon in the rat;inject0.2mL uric acid solution (25mg/ml) into the articular cavity. Active the joint to make the drug spread to each part of the joint. Inject0.2mL normal saline into the normal group’s articular cavity for normal control.After making the model successful, according to the human and animal body surface area conversion formulas, calculate the does of the xiaoyan powder,futaning cream, three times per day. Observe the dministration for three day s;kill the rats with decapitation method.Take the ankle synovium,then use immunohistochemical method under light microscope to observe the activity of membrane of intercellular adhesion molecule1(ICAM-1).Results:The analgesic experiment:Xiaoyan powder group and Futalin group could improve the pain threshold and reduce the writhing of the miceto some degree.(P>0.05), There is no obvious difference between the two groups. Compared with the normal group (p<0.05), the differences have statistical significance.Gout arthritis drug intervention experiment:The integral optical density sum of the rat knee joint synovial in the model group is more than the Xiaoyan powder group, Futalin group and normal saline (P<0.05). There is no obvious difference between the Xiaoyan powder group and the Futalin group(P>0.05); the Futalin group is less than the normal saline group (P<0.05), the differences have statistical significance.; the Xiaoyan powder group and the normal group (P<0.05), the differences have statistical significance.Conclusion:Xiaoyan powder has the anti-inflammatory analgesic effect in mice,it can also reduce the ICAM-1content of the rat knee joint synovial, and thus control the gouty arthritis, it has preventive and therapeutic effects for the the rat ankle joint gouty arthritis.
