Objectives:To detect the expression of interleukin-18(IL-18) and human cartilage glycoprotein-39(YKL-40) in serum and placenta of patients with preeclampsia and analyze their correlation in preeclampsia, then provide the theoretical basis for investigating the pathogenesis of preeclampsia.Methods:20cases of normal pregnant women and50women with preeclampsia (20cases of mild preeclampsia group;30cases of severe preeclampsia group) were selected. The expression level of IL-18and YKL-40in serum and placenta between the two groups were determined by ELISA and immunohistochemistry. All pregnant women were collected prior to labour onset, at caesarean section.There were no significant difference in the age, body mass index, gestational weeks, parity and gravid in the two groups. They were diagnosed according to the criteria (Obstetetrics and Gynecology, the Seventh Edition), without complications of internal medicine and other complications of pregnancy.Results:(1) The expression of IL-18protein was mainly observed in the endochylema of placental villous sycytiotrophoblasts and cytotrophoblast, a few in cytoplasm of interstitial cells and vascular endothelial cells and occasionally in nucelus.The expression level of IL-18in placenta in patients with preeclampsia was significantly higher than the control group (p<0.01). The expression level of IL-18in placenta in patients with severe preeclampsia was significantly higher than in patients with mild preeclampsia (p<0.01).The expression level of IL-18between patients with mild preeclampsia and control group has no significant differences (p>0.05).(2) YKL-40was mainly located in endochylema of placental villous sycytiotrophoblasts and vascular endothelial cells,a few in nucelus and endochylema of interstitial cells.The expression level of YKL-40in patients with preeclampsia was significantly higher than the control group (P<0.05). The expression level of YKL-40in patients with severe preeclampsia was significantly higher than in patients with mild preeclampsia (p<0.05).The expression level of YKL-40protein between patients with mild preeclampsia and control group has no significant differences(P>0.05).(3)According to ELISA, the expression level of IL-18and YKL-40in serum of patients with preeclampsia was significantly higher than the control group(P<0.01). Notably, higher serum YKL-40and IL-18levels were observed in patients with severe preeclampsia than in patients with mild preeclampsia(P<0.05).(4)Correlation analysis showed that the expression of IL-18and YKL-40in placenta and serum of patients with preeclampsia had a positive correlation (rt=0.567,r2=0.368,P<0.01)Conclusionsr(1) The expression level of IL-18and YKL-40protein in placenta and serum of the preeclampsia group was higher than the control group and correlated with the severity of the disease.(2) The expression of IL-18and YKL-40in placenta and serum of the preeclampsia group had a positive correlation, which may participate in the pathogenesis of preeclampsia.
