Background and ObjectivesHypertensive disorder complicating pregnancy(HDCP)is a specific sydrome to humans gestation, which may seriously impair the health of maternity and infant, in which preeclampsia is the most frequent type. The specific etiology and pathogenesis of Pre-eclampsia is still inconclusive although there have been many findings and hypotheses of the studies on them. At present placental's shallow implantation and vascular endothelial dysfunction are considered as the major doctrine of Pre-eclampsia, along with more and more evidence to this doctrine. As the special organ during pregnancy, placenta almost takes all functions for immature fetal organs. Estrogen, progesterone and oxytocin are the main gestational placenta secrete hormones which play an indispensable role during the period of gestational maternal adaptation; maintain gravid, fetal growth and childbirth startup. Some scholars have found that estrogen, progesterone and oxytocin and their receptors are involved in the protection of endothelial function of blood vessels, but its exact mechanism is unclear right now.Now there is seldom research on estrogen receptor (ER), progesterone receptor (PR) and oxytocin receptor (OTR) performance in placenta organization Pre-eclampsia patients. Through detecting the performance and their correlation analysis of ER, PR and OTR in normal late pregnant female and preeclampsia patients, this research tries to discuss their effects during the development of pre-eclampsia, further to understand the etiology and pathogenesis of preeclampsia and provides a new theory basis for early prevention, early diagnosis and the monitoring of the illness.Methods90 patients with hypertensive disorder complicating pregnancy were selected from June 2009 to March 2010 in the Second Hospital of Zhengzhou University, and the expression levels of ER, PR and OTR in the placental tissue were detected by immunohistochemistry. There were 30 cases of hypertensive disorder pregnancy,30 patients with mild preeclampsia,30 cases severe preeclampsia, and 30 cases of normal late-pregnant women (control group).Results(1) ER was mostly expressed in the nucleus of syncytiotrophoblast of placental villi, and slightly expressed in the nucleus of decidual cell and villus syncytiotrophoblast. There were statistically significant differences about the expressions of ER among gestational hypertension, mild and severe preeclampsia group. The expressions of ER in mild and severe preeclampsia group was significantly lower than that in the control group (P<0.05). (2) PR was mostly expressed in the nucleus and (or) cytoplasm of placental syncytiotrophoblast, The expressions of PR in gestational hypertension, mild and severe preeclampsia group were lower than that in the control group(P<0.05),The expressions of PR in mild and severe preeclampsia group was significantly lower than that in the control group(P<0.05). (3) OTR was mostly expressed in the membrane and cytosolic of placental syncytiotr ophoblast cells, and there were positive expressions in the cell membrane and cytoplasm of trophoblastic cells. The expressions of OTR in gestational hypertension, mild and severe preeclampsia group were significantly lower than that in the control group (P <0.05). There were significant differences about the expressions of OTR between gestational hypertension group and mild and severe preeclampsia groups. (4)Nuonatal weights and placenta weights of mild and severe preeclampsia group were significantly lower than that in the control group (P<0.05). (5) There were positive correlations about the expression levels of ER and PR between control group and hypertensive disorders in pregnancy in the placental tissue, (r=0.873, P<0.01;r=0.816P<0.01;r=0.848, P<0.01; r=0.806, P<0.01).(6) There were positive correlations about the expression levels of ER and OTR between control group and hypertensive disorders in pregnancy in the placental tissue (r=0.876, P<0.0; r=0.847, P<0.01; r r=0.857, P<0.01; r=0.859, P<0.0).(7) There were positive correlations about the expression levels of PR and OTR between control group and hypertensive disorders in pregnancy in the placental tissue (r=0.914, P<0.01; r=0.811, P<0.01; r=0.882,P<0.01;r=0.875,P<0.01)Conclusions1. ER,PR and OTR may play important role in promoting penetration of syncytiotrophoblast, placenta mature and process of fetal growth.2. The reduction about the expression level of ER, PR and OTR in pre-eclampsia may have effect on the pathogenesy of pre-eclampsia...
