Objective:Qingjie Ligan mixture is a kind of traditional Chinese medicine, which is made from forsythia, honeysuckle, pardanthus and mint, with the function of clearing away heat and toxic material, clearing throat, and expelling pathogenic wind from the body surface. On the basis of production process feasible and quality standards controllable, tests of antipyretic and analgesic, anti-inflammatory, immune, and activity of antibacterial anti-viral was carrying out to supply pharmacodynamic experimental basis for the declaration of the clinical documents of approval of the Chinese medicine, natural medicine and new drugs.Methods:Rats febril models induced by LPS and yeast was used, via determining changes in body temperature and levels of NO and TNF-alpha,5-HT in serum to investigate the antipyretic efficacy of Qingjie Ligan mixture. The permeability change of celia capillary auricle swelling degree, mouse paw edema caused by carrageenin, acetic acid tests and resistar rate caused by dimthylphenanthrene were taken as indexes to analyze the clearing throat function of the mixture. The clearance rate of charcoal particles,production of serum hemolysin and delayed hypersensitivity reaction of this mixture were used as indexes to the function of expelling pathogenic wind from the body surface.In vitro anti-respiratory tract bacteria, mice, anti-alpha hemolytic streptococcus and Staphyl and Anti-alpha hemolytic streptococcus and Staphylococcus aureus test in vitro resistance to the HlNl, RSV, HSV-1, CVB3and mice against HlNl influenza virus test were investigated to clear solutions facilitate a sense of detoxifying effect of the mixtureResults:1.The dose of2,4,8,16ml/kg of Qingjie Ligan mixture can inhibit heat generation induced by LPS and yeast after gavagied continuously for3days, TNF-a and NO levels in serum of the model animalare were reduced to varying degrees, but significantly enhance5-HT levels of serumin yeast fever model,but no significant effect on LPS fever model.The dose of2.5,5,10,20ml/kg of Qingjie Ligan mixture could significantly inhibit xylene-induced ear edema and capillary permeabilifility increase in mice, hind paw edema induced with carrageen and granuloma induced with cotton ball in mice, Evans blue excretion of mouse peritonitis induced by acetic acid. And this mature can easy acetic acid tests to some degree.3. The dose of2.5,5,10,20ml/kg of Qingjie Ligan mixture can significantly improve the prednisone acetate immunogenic mice with low carbon clearance index K and the thymus index, increase phagocytic coefficient a and spleen index to Varying degrees.It can significantly improve the immunization of mice with low serum hemolysin levels and thymus and spleen index, specific humoral immunity in normal mice had no significant effect.It also can enhanced acetate prednisone induced immunocompromised mice delayed-type hypersensitivity, have varying degrees of inhibition of specific cellular immunity in normal mice to vary degrees.4. The results in vitro show that the Qingjie Ligan mixture can significantly inhibite the common pathogens of respiratory tract infection by alpha-hemolytic Streptococcus, Mycobacterium tuberculosis, Sarcina lutea, Staphylococcus aureus, beta hemolytic Streptococcus, Staphylococcus epidermidis were most sensitive. Influenza A virus (H1N1), respiratory syncytial virus (RSV), herpes simplex virus type1(HSV-1) were inhibited, wih the IC501/2560,1/2560,1/1280, but this mixture has no significant inhibitory of Coxsackie virus group B type3(CVB3).5. The results of test in vivo show that10,20,40times the human dosage of the Qingjie Ligan mixture in the experimental mice showed anti-influenza virus infection model in mice, the average survival days is prolonged from6.63days to9.50and11.30days, extend the life xate from43.29%to70.44%, and increase death protection rate from36.67%to56.67%. The minimum markedly dose is10.40g/kg and the therapeutic index is8.0. This mixture can antagonize infection of mouse by alpha hemolytic streptococcus, the ED50was36.99mg/kg.bw, but had no significant protective effect for aureus infected mice.Conclusion:The Qingjie Ligan mixture in vitro and in vivo could significantly inhibit the respiratory tract pathogenic bacteria and viruses, reduced body temperature of rat model induced by both bacterial, suggesting its detoxification effect.significantly inhibited in mice with acute, subacute and chronic inflammation, writhing pain inhibition, suggesting that its throat clearing effect is clear. It also could significantly enhance the immunization of mice with low non-specific and specific humoral immunity and cellular immunity, but it has little effect to normal mice function, suggesting that its can expelling pathogenic wind from the body surface. All of the results provide an experimental evidence for the clinical application of Qingjie Ligan mixture.
