The Shatian hospital used Major decoction ofgreen dragon to treate influenza patients, efficacy significantly, especially inthe antipyretic effect. On the basis of pre-clinical efficacy, this thesis systemstudied antipyretic and anti-Influenza A H1N1influenza virus of the Majordecoction of green dragon, whose purpose is to interpret itspharmacodynamic constituents, and provide a scientific basis for the clinicalefficacy of the classic recipe. This thesis reviewed the titerature about Major decoction of greendragon and anti-influenza virus and antipyretic at home and abroad on theChinese systematically,summed up the classic recipe evidence of sidesolution, prescription smell chemical constituents and pharmacologicaleffects and clinical applications, as well as traditional Chinese medicineantipyretic and anti-influenzamore systematic review of the effects of thevirus research profile, and provided a reference for subject design.Major decoction of green dragon considered as a whole, by supercriticalCO2extraction method and system solvent is decomposed into parts ofdifferent chemicals such as supercritical CO2extract (volatile oil), totalpolysaccharides, alkaloids, phenolic acids,glycosides, raffinate aqueousphase and gypsum decoction. Pharmacological screening of experimentalstudies have shown the supercritical CO2extract and glycosides has obviousanti-influenza AH1N1virus activity; the alkaloids, polysaccharides andplaster antipyretic obviously. The chemical composition of extract fromRamulus Cinnamomi-Zingiber Recens is analysised by GC-MS,55compounds were identified, and the relative content of the total compositionis97.29%. the main components are an aldehyde, ketone, ester compounds.The main active ingredient of cinnamic aldehyde relative content62.85%,the2-methoxycinnamaldehyde16.03%; ginger (+)–aromadendrene2.34%,6-gingerol to0.37%, alpha-zingiberene0.14%; The n-butanol extractingredients parts,another anti-influenza virus material, was analysised byHPLC-MS,identified22of the35compounds, such as Amygdalin,Rutin,Liquiritin and Glycyrrhizic acid ammonium salt.Many compoundsderives from Radix glycyrrhizae preparata,and aglycone structure is complex,including flavonoids, terpenes. The background of Major decoction of greendragon anti-influenza virus substance is explained by the study. This article researched the efficacy and its mechanism of actioneffective of substances fraction of anti-influenza virus from Major decoctionof green dragon.Intranasal infection Babl/c mice with H1N1influenza virusadapted strains of mice lung (FM/1/47strains), gavage the antiviralsubstances from the decoction, comparison decoction effect and explore itsmechanism. Pharmacodynamic studies, including the protection ofexperimental drug to be tested on influenza virus induced death, druginfluenza virus infection in mice with viral pneumonia. Evaluation indexdeath rate of protection, spleen index, thymus index, lung index, theinhibition rate of lung lesions, lung tissue structural changes. The mainmechanism of anti-influenza virus drugs against influenza virus infection inimmune factors in serum and lung tissue, evaluation index of inflammatorycytokines, Inflammatory factors TNF-α,IL-6;Antiviral factor IFN-γ; andanti-inflammatory cytokine IL-10.The results show that Major decoction ofgreen dragon antiviral substances fraction can significantly prolong thesurvival time of mice infected with influenza virus, significantly inhibit viralpneumonia. But the the role that reduced the immune organs, spleen andthymus weight and organ index, suggests that the decoction may be inhibitedimmune organs, so clinical use to avoid excessive and long time. Theantiviral material fraction anti-influenza virus reduces the weight of immuneorgans stronger than the decoction. The infection mice serum TNF-α, IFN-γ, IL-10significantly reduced, IL-6significantly increased; lung tissues inthe TNF-α, IFN-γ and IL-6were significantly elevated, IL-10wassignificantly reduced.The immune system of the infection mice appearsserious disorder, specific manifestations of systemic immune suppression,local lesions of the lungs excessive immune.The decoction anti-influenzavirus fraction of effective substances in the serum TNF-α, IFN-γand IL-10increased, reducing the role of IL-6,correct inflammatory factorsover expression in the body, thereby reducing systemic inflammatory response,lowering effect in the lungs of mice TNF-α, IFN-γ.slightly elevated IL-6,IL-10, a slight effect. The serum TNF-α, IL-1βof drug-treated ratssignificantly reduced modeling after2hours; and modeling after4hours,thecAMP of hypothalamus significantly reduced;Rats modeling after4hours,the calcium of hypothalamus significantly higher in the low dose andgypsum low and high group,the high-dose group and the all-party group alsotended to increase, but has no significant difference. The antipyreticmechanism of the soup and antipyretic effective substances fraction is toreduce peripheral pyrogenic factor TNF-α, IL-1β, reduce the cAMP inhypothalamus and increased the calciumin hypothalamus.Combine the anti-influenza virus and antipyretic active substances group parties, to explore the role of anti-influenza virus in joint group side and anti-influenza virus the effective substances fraction side,to explore the roleof antipyretic effect in the combined group and effective antipyretic materialparts prescription.The results show that the side of the combined groupshowed better anti-influenza virus and antipyretic effects,superior efficacysubstances fraction effect, so we can say that synergy is in the anti-influenzavirus and antipyretic material part; the antiviral material parts can enhanceantipyretic effect, demonstrate the synergy of two pharmacodynamicmaterial parts. Studies have shown that anti-influenza virus material partsand antipyretic active substances parts synergie. The research hasaccumulated useful experience on the component Chinese medicine,provises modern scientific basis for compatibility synergy theory and theinterpretation of Chinese medicine.
