Infectious hyperpyrexia belong to Chinese medicalemergency.The mean clinical symptoms are high fever (temperture>39℃),morbid thirst,rapid pulse.The disease are same asemergency infectious hyperpyrexia ,chronic disease superventinfertious hyperpyrexia,nervous system central hyperpyrexia and soon.Infectious hyperpyrexia have some peculiarties such as: invasionrapided,multifarious noxa,involved nosogenesis,progress quicklyand converst easily.At persent,modern medicine therapy it withantipyretic,analgesic,antiviral and preventive infection.But that cannot block the development of the disease,the side effects are mightalso.Chinese medicine studied the disease thousands of yearsago,and have accumulated plentiful experiences.We can therapy itwith syndrome differ entiation.The effectiveness is very great andside effects are much small.Beause that,on the basis of RGQprescription made by famous old practition of Chinese medicine GaoShengsan ,we design and make the preparation plan the experimentto research how to cure infectious hyperpyrexia and proved someexperiment prove.Objective:Through five experiments,we attempt to study theRGQ pharmacodynamic mechanism.The five experiments are feverresulted from saccharomyces,turgescence resulted fromdimethylbenzene,pain resulted from acetic,antibacterial in vitro andantiviral in vitro.Methods:Experiment one:RGQ effect the fever resulted fromsaccharomyces.50 Wistar rats,male,weight 190-220g,life in laboratory for fivedays.Mcasure the temperature of anus in the after three days.Before12 hours of the experiment,the rats can drink water freely,but can noteat anything.On the morning of the experiment we mcasure thetemperature of anus with two times and make the mean temperatureas the normal.We inject 20% suspension of saccharomyces 10 ml/Kginto the back of the big rat.After fevering,we divide rats into fivegroups at radom.The five groups are RGQ13.6g/Kg,6.8 g/Kg,3.4g/Kg group,aspirin 100mg/Kg group and model group.we record thechange of temperature at the time of 1,2,4,6and 8hour.Experiment two:RGQ effect the ear's turgescence resultedfrom dimethylbenzene.50 Knuming rats,male,weight 28±2g.Divide them into fivegroups at random.They are modle group,aspirin 100mg/Kggroup,RGQ 20.0g/Kg,10.0 g/Kg,5.0 g/Kg group.Irrigate RGQ torats on the continuous three days.At the first hour after the lastirrigten,we smear dimethylbenzene 0.1ml on the rate's right ear'stwo sides.After four hours,we sacrifice them and shear two ears ofevery rate on the auricle.We record the difference of two ear'sweight as the turgescence degree.Experiment three:RGQ effect the pain resulted from acetic.50 Kunming rats,The method of divide groups and irrigate drugare same as experiment two.At the second hour after irrigateRGQ,we inject 0.5% acetic 0.1ml/10g into rate's celiac.Watch themhow many contortions in 10 minutes.Experiment four:Antibacterial in vitro.Make filter paper into ring-shaped and take then intodehumidifier after autoclave.Dilute RGQ with distilled water as1.5g/ml,0.75 g/ml,0.325 g/ml,0.17g/ml respectively.Immerse filterpaper into different density RGQ for 30 mintues and take them into37℃ thermostant.We make colomy into bacteria suspension aftercultured in nutrient medium.Take 1ml bacteria suspension ontonutrient medium and make it dry.Under the aseptic technique,putdifferent density filter paper on the surface of bacteria medium.Takethem into 37℃ thermostant for 18 hours.Experiment five:Antiviral in vitro.Digest HEp-2 with the methord of cell passage when the HEp-2begin exuvia.We divergence HEp-2 with culture.Dilute HEp-2 as 1.5×105 cell/ml,put 100ul cell into 96 microdose culture panel.Dilutevirus suspension with EMEM at ten times reduction.Inoculatdifferent density virus into 96 microdose culture panel whenmonodermal have grouped up.Watch the change of cell under themicroscope and calculate TCID50.Basised on the method of ReedMuench,calculate the titration of virus.At last,acquisite 30TCID50.Mix different density RGQ 100ul into 96 microdose culturepanel when monodermal have grouped up and watch the change ofcell under the microscope,calculate TCO0 and TCO50.Inoculate 30TCID50 into HEp-2 96 microdose culture panel that divide viruscontrol,RGQ control,postive(ribavirion)control and cell control.Put itinto 37℃ 5% CO2 culture for three days.Everyday watch and recordthe change of cell under microscope.Results:Experiment one:Calculate with t text,at the time of 1,2,4,6and 8 hour,the comparison of therapied group and model group is p<0.05.Compare RGQ group and aspirin group p>0.05.Experiment two:The comparison of RGQ high and middledose group and model group is p<0.05.RGQ have the effect ofanti-inflammatory.But compare RGQ low dose group and modelgroup is p>0.05.Compare RGQ group and aspirin group p>0.05.Experiment three:The RGQ high,middle,low dose group caninhibit the pain reaction brazenly such as aspirin.Calculat with ttext,compare the therapied group and model group is p <0.05.Compare RGQ and aspirin group p>0.05.Experiment four:RGQ can inhibit Escherichia coli, pneumo-coccus,Staphycococcus aureus,Haemophilus influenzae, Bacteriumand Flexner's baccillus on the different degree and the effect ofinhibittion can be inharm when the density have been increased.Experiment five:On the conclution that infected 30TCID50,RGQ can inhibit echovirusⅡ,Influenzavirus FM1, adeno-virus3 and can delay herpesvirusⅠalso.Conclusion:On the base of the experiment's results,RGQ havethe effection that conclude antipyrectic, analgesia, anti-b inflamm-atory,antibacter and antiviral.Discussion:Chinese medicine think that infectious hyperpyrexia is fevered because that six exopathogens and pestilence whoattack stria connecting tissue sturggle between healthy qi andpathogen.The cause of disease is apoplexied seasonal pathogenicfactors.The body is disordered of wei qi.The pathogenesis arestruggled between healty qi and pathogen.The mean syndromes arewind-cold syndrome,wind-heat syndrome,summer hygrosis orpyretic pulmonary syndrome.Chinese medical center is theapied withsyndrome differ entiation,cure it through different means.Themethod isn't anti-inflammatory and antipyretic simply,but the aim isthat enhance the power of immune system and defence disease.Thestudy of moden pharmacology proved that Chinese medicine ofdiaphoresis relieving superficies prescription have the effect ofantipyretic and regulated immune system.RGQ maked up strictlyhave the impact of diaphoresis relieving superficies,cooling qi fenpurgation and releasing orifices.RGQ can cure infecturehyperpyrexia safely and effictively and be good at clinic using.
